跑步运动对去睾大鼠腰椎骨量及骨结构的影响

摘  要:目的:探讨不同强度的跑步运动对去睾大鼠非负重骨腰椎的骨量和骨显微结构的影响和机制。方法:12周龄SD雄性大鼠40只,随机分为5组,即假手术组(SHAM)、去睾模型组(ORX)、去睾后低强度运动组(ORX+EX1)、去睾后中强度运动组(ORX+EX2)、去睾后高强度运动组(ORX+EX3)。12周后,处死取腰椎骨,做不脱钙骨切片,并测定骨形态计量学各骨量、骨结构、骨代谢参数。各组参数作方差分析,显著性检验。结果:(1)ORX组各参数与SHAM组比较显示:骨量降低(P<0.05),骨显微结构破坏(P<0.05),成骨减弱(P<0.05),破骨活跃(P<0.05)。(2)ORX+EX1和ORX+EX2组各参数与ORX组比较显示:骨量增高(P<0.05),骨显微结构改善(P<0.05),成骨活跃(P<0.05),破骨减弱(P<0.05)。(3)ORX+EX3组各参数与ORX组比较显示:骨量和骨结构均无明显改善(P>0.05),但成骨和破骨均活跃,骨转换加快(P<0.05)。结论:(1)雄性大鼠去睾后能引起骨量丢失及骨结构破坏。(2)低中强度的跑步运动对去睾大鼠不仅能增加骨量还能改善骨显微结构。(3)高强度的跑步运动不能减缓去睾大鼠骨量丢失和骨结构破坏。Objective:This study explored the effect and mechanism of different intensity tread mill exercise on nonweight-bearing bone mass of the orchidectomized Rats.Methods:Forty male Sprague Dawly rats aged 12 weeks were divided into following groups: SHAM, ORX, three groups of individual exercise of different intensity: ORX+ EX1,ORX+ EX2 and ORX+ EX3. After running for 12 weeks, the rats were sacrificed and the 4th lumbar vertebral body (L4) was collected to make bone slices without decalcification, meanwhile, to determinate diverse indexes about bone mass, bone architecture, bone metabolism and bone morphological metrology.Results:(1) Compared with SHAM group, bone mass in ORX groups was significantly decreased (P0.05), bone architecture was destroyed (P0.05), with weakened ossification (P0.05) and increased osteoclast (P0.05). (2) Compared with ORX group, bone mass in ORX+EX1 and ORX+EX2 groups was obviously increased (P0.05), with improved bone architecture, increased ossification and weakened osteoclast (P0.05). (3) Compared with ORX group, bone mass and bone architecture in ORX+ EX3 group showed no remarkable changes (P0.05), but ossification and osteoclast were both activated, and bone turnover was accelerated (P0.05).Conclusion:(1) Orchidectomy will cause bone mass and destroy bone structure. (2) Low and medium intensity running can increase bone mass and improve bone architecture. (3) Excessive running exercise cannot maintain bone mass and bone architecture.
