摘 要:目的研究基底拉伸应变对小鼠成骨细胞系MC3T3-E1、破骨细胞系RAW264.7及骨细胞MLO-Y4三种细胞BMP-2mRNA表达的影响。方法三种细胞随机分为0με、1000με、1500με、2000με、2500με和5000με组,最佳拉伸时间和周期为1次/d,每次1h,连续3d,频率为0.5Hz。采用卫生装备研究所自行设计研制的四点弯曲装置对小鼠三种细胞进行拉伸加载。采用RT-PCR技术分别研究不同应变对小鼠三种细胞BMP-2mRNA表达。结果 MC3T3-E1细胞RT-PCR结果显示:1500με、2000με组和2500με组与0με组相比BMP-2mRNA表达显著增强(P<0.01);5000με组与0με组相比BMP-2mRNA表达显著降低(P<0.01);RAW264.7细胞RT-PCR结果显示:1500με、2000με组和2500με组与0με组相比BMP-2mRNA表达显著降低(P<0.01);5000με组与0με组相比BMP-2mRNA表达显著降低(P<0.01);MLO-Y4细胞BMP2基因表达结果与MC3T3-E1一致。结论①BMP-2在成骨细胞系MC3T3-E1、破骨细胞系RAW-264.7及骨细胞系MLO-Y4三种细胞中均有表达;②1500με、2000με、2500με三种生理剂量的拉伸应变可以显著增加MC3T3-E1、MLO-Y4细胞BMP-2的表达,并呈剂量依赖性,超生理剂量5000με可以显著降低MC3T3-E1、MLO-Y4细胞BMP-2的表达;③相同的力学拉伸作用条件下,BMP-2在RAW-264.7细胞中表达与MC3T3-E1、MLO-Y4细胞的表达趋势相反。Objective To study the effects of substrate-stretching strain on the BMP-2 mRNA expression in MC3T3-E1,RAW264.7 and MLO-Y4 three mouse cell lines.Methods Three mouse cell lines were randomly divided into six groups according to stretching cycle.They were selected once a day,once 1 h,on successive 3 days as the stretching time and 0.5 Hz as the frequency.Cells were stretched by a self-developed four-point bending loading device.The expression of BMP-2 mRNA was examined by RT-PCR.Results The BMP-2 mRNA expressions of MC3T3-E1 of 1 500,2 000 and 2 500 με groups were significantly upregulated(P0.01),and its expression in 5 000 με group were significantly down regulated comparing with that of 0 με(P0.01).The BMP-2 mRNA expressions of RAW264.7 of 1 500,2 000 and 2 500 με groups were significantly downregulated (P0.01),and its expression in 5 000 με group was significantly upregulated comparing with that of 0 με(P0.01).BMP-2 mRNA expression of MLO-Y4 was similar with MC3T3-E1's.Conclusions (1)BMP-2 can be expressed in MC3T3-E1,RAW264.7 and MLO-Y4 three mouse cell lines.(2) Substrate-stretching strain of physiological dose(1 500,2 000,2 500 με) can significantly upregulate the expression of BMP-2 mRNA in a dose-dependent manner in MC3T3-E1,MLO-Y4 cell lines.Substrate-stretching strain of overphysiological dose(5 000 με) can significantly downregulate the expression of BMP-2 mRNA in these two cell lines.(3) The tendency of BMP-2 mRNA expression in RAW-264 cells is opposite with MC3T3-E1,MLO-Y4 at same condition.
