摘  要:目的探讨巢式酶链聚合反应(PCR)技术快速检测泌尿生殖道致病性支原体在临床上的应用。方法以支原体16S^23S保守区域基因为扩增靶序列设计通用引物,采用巢式PCR方法扩增微小脲原体(Up)、解脲脲原体(Uu)、生殖支原体(Mg)和人型支原体(Mh)4种支原体。结果巢式PCR检测灵敏度高于一步PCR(2χ=5.857,P<0.05),巢式PCR扩增出4种支原体的标准株,对1份生物样品可同时检测出4种致病性支原体,避免了误诊、漏诊,提高诊治效率。结论巢式PCR技术能快速、敏感、准确检测出Up、Uu、Mg和Mh引起的单一感染和混合感染,对于临床感染性尿道炎的早期诊断有重要意义。Objective To evaluate the clinical application of nested polymerase chain reaction(PCR) for the fast detection of infective mycoplasma in genitourinary tract.Methods The four mycoplasma ureaplasma parvum(Up),ureaplasma urealyticum(Uu),mycoplasma genitalium(Mg) and mycoplasma hominis(Mh) were amplified by nested PCR with consensus primer which was designed according to 16S-23S conservative region gene of mycoplasma.Results The sensitivity of nested PCR was higher than that of one step PCR(χ2=5.857,P0.05),and four types of mycoplasma were amplified by nested PCR.Four mycoplasma can be detected in one biological specimen,without diagnostic errors and omission,and the diagnosis and treatment efficiency was improved.Conclusions Single infection and polyinfection caused by Up,Uu,Mg and Mh can be detected by nested PCR quickly,sensitively and accurately,and it is important to diagnose clinical infective urethritis early.
