益气养阴法及其组方抗细胞衰老的实验研究

摘　　要：目的:研究益气养阴法及其组方对体外培养的人胚肺二倍体成纤维细胞衰老的影响。方法:将健康雄性SD大鼠随机分为中药组、维生素E组、生理盐水组,采用益气养阴中药、维生素E胶丸、生理盐水分别灌胃3组大鼠,提取含药血清,将提取的血清培养二倍体人胚肺成纤维细胞(MRC-5)。从第28代开始在培养液中加入10%含药血清培养细胞,观察细胞形态、细胞群体倍增曲线、细胞传代次数、细胞生长曲线情况,并进行组间对比分析。结果:中药组与维生素E组及生理盐水组比较,中药组细胞开始出现衰老的时间及细胞死亡的时间明显延迟,差异有统计学意义(P<0.01);用含药血清培养后,中药组细胞密度较维生素E组及生理盐水组明显增高(P<0.01);中药组细胞传代代数与维生素E组比较差异有统计学意义(P<0.05);而生理盐水组细胞传代代数,明显低于中药组及维生素E组(P<0.01)。结论:益气养阴中药组方可以显著延长体外培养的人胚肺二倍体成纤维细胞的寿命,同时能增强衰老细胞的增殖分裂能力,从而延缓细胞的衰老。Objective：To explore the effect of Tonifying-qi and Nourishing-yin method on fibroblast aging.Methods：Healthy male SD rats were randomly divided into TCM group with tonifying-qi and nourishing-yin herbs,vitamin E group with vitamin E and saline group with saline.The serum was extracted to culture human diploid fibroblasts（MRC-5）.The 10% serum was added into culture solution from the 28th generation.Cell shape,multiplication curve of cell colony,passage frequency,and growth curve were observed and compared.Results：The aging time and death time delayed,and the cell density increased significantly in TCM group,comparing with vitamin E group and saline group（P0.01）.There was significant difference in passage frequency between TCM group and vitamin E group（P0.05）,and the frequency was remarkably lower in saline group（P0.01）.Conclusions：Tonifying-qi and Nourishing-yin formula can obviously prolong life of human diploid fibroblasts,and strengthen multiplicative division of aging cells to delay cell aging.
