桑银降糖胶囊在体外对猪胰岛细胞增殖及分泌功能的影响

摘　　要：目的观察桑银降糖胶囊对体外培养猪胰岛细胞分泌功能的影响。方法体外分离纯化猪胰岛细胞进行单层细胞培养,链脲霉素(STZ)处理后分别与桑银降糖胶囊及其组分桑叶多糖、牛磺酸、银杏黄酮等共同孵育24、48 h,用放射免疫法检测培养液中胰岛素、C肽水平;光镜下观察细胞形态变化;MTT法检测胰岛细胞增殖情况。结果与其他组分比较,桑银降糖胶囊可明显提高培养液中胰岛素、C肽水平,胰岛β细胞数目明显增多。结论桑银降糖胶囊可减轻STZ对胰岛β细胞的损伤,促进胰岛细胞增殖,并使损伤的胰腺恢复正常功能,增强胰岛素的分泌功能。Objective To investigate the effects of Sangyin capsule on proliferation and secretion function of porcine islet cells in vitro.Methods Islet cells of new-born swine were separated for monolayer cell culture,after the performance of streptozotocin,the islet cells were treated with Sangyin capsule and its components including mulberry polysaccharide,taurine,and ginkgetin,respectively for 24 hs and 48 hs,morphology of islet cells in every group was observed with microscopy and the proliferation was determined by MTT assay.Insulin level and C peptide level in the cell culture media were measured by radioimmunoassays.Results Compared with its component groups,Sangyin capsule obviously increased insulin level and C peptide level in the cell culture media,and also increased the cell number of islets.Conclusions Sangyin capsule has the function of repairing and protecting the damaged β cells in islets of pancreas,thus to promote the proliferation and ameliorate the function of islet β cells.
