摘  要:目的:应用基因微阵列技术检测子痫前期(pre-eclampsia,PE)患者和正常妊娠胎盘组织差异表达基因,寻找PE相关发病基因,为研究子痫前期病因提供分子水平理论基础。方法:从正常妊娠和子痫前期胎盘组织中抽提mRNA,反转录后,探针标记cDNA。以混合的6例正常胎盘总RNA为对照,应用cDNA微阵列人类全基因表达谱分析芯片检测6例子痫前期患者胎盘基因表达的改变。结果:在芯片杂交过程中,有91个基因表达差异具有统计学意义,与正常对照相比,子痫前期样品中有19个基因表达下调,72个基因表达上调,这些基因涉及信号传导、运输、转录调控、细胞增殖和凋亡、脂代谢、钙离子结合以及免疫等方面的功能。结论:子痫前期发病涉及了多方面细胞功能基因表达的变化,应用cDNA表达谱芯片可快捷、高通量地筛选出子痫前期胎盘可能的致病基因,为研究子痫前期的病因提供新的理论基础。Objective: To investigate new clue for the research of the etiology of pregnancy-induced hypertension (PIH) by testing the gene expression difference between pre eclamptic placentas and normal ones. Methods: mRNA level of 6 PIH placentas were examined using 4 000 feature cDNA. Microarray were in comparison with the pooled control consisting of total RNA from 6 cases of PIH placentas after the control eDNA and experimental eDNA were labeled by cy3 and cy5 respectively. Results: A total of 91 differently expressed genes were identified in placental tissue of PIH. Seventy-two were up-regulated and 19 down-regulated. These included some ge nal and transferring proteins. Conclusions nitor the altered gene expression and the h , cellular sig method to mo for the etiologiresult could provide interesting clue and strategy cal research of PIH.
