血必净注射液对脂多糖性肝损伤大鼠PPAR-γ基因表达及肝损伤的影响

摘　　要：目的研究血必净注射液对急性肝损伤大鼠肝组织中过氧化物酶体增殖物激活受体γ(PPAR-γ)表达的动态影响。方法将SD大鼠随机分为对照组、模型组和治疗组,采用腹腔注射D-氨基半乳糖(D-Gal)、脂多糖建立急性肝损伤大鼠模型。治疗组在造模前6 h及制模后大鼠腹腔内给予血必净注射液2.5 g/kg,12 h 1次,共4次,各组于造模后6、12、24、48 h 4个时间点留取大鼠血及肝脏标本。观察大鼠肝功能及肝脏病理变化;用RT-PCR法测定各组肝PPAR-γmRNA表达水平;ELISA法测定血清中肿瘤坏死因子-α(TNF-α),白细胞介素-6(IL-6)水平。结果模型组大鼠肝组织PPAR-γmRNA表达明显低于对照组,治疗组组织大鼠肝PPAR-γmRNA水平明显高于模型组,组间比较差异有统计学意义。结论血必净注射液对实验性急性肝损伤有一定保护作用,其机制可能与升高鼠肝PPAR-γmRNA表达水平有关。Objective To investgate the effect of Xuebijing on peroxisome proliferators-activated receptor γ（PPAR-γ） in the liver of the acute liver injury model rats induced by endotoxin.Methods One hundred and eighty SD rats were divided into 3 groups randomly：control group,model group and treatment group.The model of acute liver injury was induced by injecting lipopolysaccharide and D-galactosamine（D-Gal） into abdomen.Xuebijing was injected into the abdomen of the rats in treatment group,and at 6 h before the establishment of models and 12,24,36 h after the establishment of models injected into the other side abdomen.At 6,12,24,48 h after the establishment of models,serum and liver samples were collected from rats to determine the levels of tumor necrosis factor-α（TNF-α）and interleukin-6（IL-6） by ELISA,and PPAR-γ mRNA by RT-PCR.Pathologic change of liver tissue was examined by microscope.Results The levels of IL-6 and TNF-α of serum in treatment group were obviously reduced than those in model group（P〈0.05）;The level of PPAR-γ mRNA of liver tissue in treatment group was higher than that in model group（P〈0.05）.Conclusion Xuebijing has a protective effect through the way of promoting PPAR-γ mRNA level to the acute liver injury model rats induced by endotoxin.
