新风胶囊对佐剂性关节炎大鼠心功能及血清细胞因子、调节T细胞的影响

摘　　要：目的观察新风胶囊对佐剂性关节炎(AA)大鼠心功能及血清细胞因子、调节T细胞的影响。方法将60只大鼠随机分为健康对照组(NC)、模型对照组(MC)、甲氨喋呤(MTX)组、雷公藤多苷片(TGT)组和新风胶囊(XFC)组,每组12只,分别向除健康对照组以外的其余动物右后足跖皮内注射弗氏完全佐剂0.1 ml致炎,致炎后第19天开始给药。健康对照组及模型对照组均给予0.9%氯化钠注射溶液,其余三组分别给予MTX、TGT、XFC。观察各组大鼠足跖肿胀度(E)、关节炎指数(AI)、心功能、血清细胞因子(TNF-α、IL-17、BNP、IL-10)、调节T细胞(CD4+Treg,CD4+CD25+Treg)的变化。结果与MC组比较,XFC组HI、LVSP和LVEDP显著降低(P<0.05),+dp/dtmax显著升高(P<0.05);与XFC组比较,MTX组体质量显著降低(P<0.01),E、AI、LVSP显著升高(P<0.05),+dp/dtmax显著降低(P<0.05)。与MC组比较,XFC组TNF-α、IL-17、BNP显著降低(P<0.05或P<0.01),IL-10、CD4+Treg、CD4+CD25+Treg显著升高(P<0.01);XFC组与其余两治疗组无显著差异(P>0.05)。结论 AA大鼠存在不同程度的心功能变化。新风胶囊在改善AA大鼠足跖肿胀度、关节炎指数的同时,也能改善其心功能,其机制可能与新风胶囊抑制异常炎症免疫反应有关。Objective To study the influence of Xinfeng capsule（XFC） on the cardiac function,regulate T cells（Treg） and cytokines（TNF-α,IL-17,BNP,IL-10）in adjuvant arthritis（AA） rats.Method 60 rats were averagely divided into normal control group（NC）,model control group（MC）,methotrexate（MTX）treated group,tripterygium glycosides tablet（TGT） treated group,XFC treated group,12 in each.Except for the rats of normal group,the others were intracutaneously injected with 0.1 ml of Freund＇s complete adjuvant（FCA） in the right hindlimb.After 19 days,NC group and MC were given normal saline,the other three groups were given MTX,TGT,XFC.All of the indexes were investigated in different groups.Results 1.When compared with the MC group,the LVSP,LVEDP（P〈0.05）and ±dp/dtmax of XFC group were significantly lower（P〈0.05）.When compared with the XFC group,the LVSP,LVEDP MTX of group were significantly higher（P〈0.05）.However the±dp/dtmax of MTX group was significantly lower（P〈0.05）.2.When compared with the XFC group,the TNF-α、IL-17、BNP were lower than that of MC group（P〈0.01）.However the IL-10,CD4^＋Treg and CD4^＋CD25^＋Treg were higher than that it（P〈0.01）.But there were no obvious differences between the two treated groups（P〉0.05）.Conclusion There were cardiac functional impairment in AA rats.Not only XFC can improve paw swelling and arthritis index,but also improve cardiac function of AA rats,which may be related to its role in regulating the abnormal immune respouses.XFC can regulate the immune response while inhibite abnormal inflammatory of immune responses which could damage myocardial cell.
