摘 要:背景高血压是最常见的心血管疾病,而高血压实验动物模型在高血压研究中扮演着重要角色。目的建立稳定的甲状腺功能亢进(甲亢)性高血压家兔模型,观察其相关指标的动态变化。方法选用新西兰大耳白家兔,给家兔皮下以0.5mg/(kg·d)的剂量分别注射L甲状腺素(T4)3、6、12和14d,制备不同程度的甲亢性高血压家兔模型,并观察心脏质量指数、体温、心率、收缩压、血浆心房利钠肽(ANP)水平的动态变化。结果 T4注射12d组存活率明显高于14d组。T4注射3d后,各项指标如心脏质量指数、体温、心率、收缩压和血浆ANP水平较正常对照组升高,其增高幅度分别为13.9%、1.4%、33.5%、11.9%和37.5%;T4注射12d组与14d组相比,上述指标无明显差异,表明12d组则进入较稳定的阶段,此时各项指标较正常对照组升高,其增高幅度分别为67.0%、3.3%、57.9%、29.3%和365.9%。单因素相关分析显示,在甲亢性高血压造模过程中心率、收缩压和血浆ANP水平与心脏质量指数之间均呈正相关(心率与心脏质量指数:r=0.927,P<0.01;收缩压与心脏质量指数:r=0.976,P<0.01;血浆ANP水平与心脏质量指数r=0.824,P<0.01)。结论给家兔皮下以0.5mg/(kg·d)的剂量T4注射12d,可获得较稳定的甲亢性高血压家兔实验模型,且在甲亢性高血压造模过程中心率、收缩压和血浆ANP水平与心脏质量指数之间均呈正相关。此造模方法较简便、成功率高、周期较短和经济易行。Background Hypertension is one of the most common cardiovascular disease,and hypertensive animal models play an important role in its experimental study.Objective To establish an animal model of thyroxine-induced hypertension and dynamic changes in related parameters.Methods New Zealand White rabbits were used in this evaluation study.Various digree of hyperthyroid models were induced by administering L-thyroxine (T4,0.5 mg/kg,sc) daily for 3,6,12 and 14 d.Cardiac mass index,body temperature,heart rate,systolic blood pressure and the levels of plasma atrial natriuretic peptide(ANP)were observed.Results The mortality rate of thyroid hormone injection for 12 d was significantly lower than that of 14 d.Thyroid hormone injection for 3 d showed characteristic changes in cardiac mass index,body temperature,heart rate,systolic blood pressure and the levels of plasma ANP,which were significantly increased by 13.9%,1.4%,33.5%,11.9%,and 37.5% compared with the control group.No significant changes of parameters was observed between 12 d and 14 d groups.Thyroid hormone injection for 12 d showed stable dynamic changes of the parameters.Cardiac mass index,body temperature,systolic blood pressure,heart rate and the levels of plasma ANP were significantly increased by 67.0%,3.3%,57.9%,29.3%,and 365.9% compared with the control group.A univariate linear regression analysis showed that heart rate,systolic blood pressure and the levels of plasma ANP were positively correlated with cardiac mass index (heart rate vs cardiac mass index:r=0.927,P〈0.01; systolic blood pressure vs cardiac mass index:r=0.976,P〈0.01; the levels of plasma ANP vs cardiac mass index:r=0.824,P〈0.01).Conclusion A stable model of thyroxine-induced hypertension rabbit model can be established by subcutaneously daily injection of T4(0.5 mg/kg)for 12 d.Heart rate,systolic blood pressure and the levels of plasma ANP were positively correlated with cardiac mass index in this model,which could be easily induced in short time of a lower cost
