摘 要:目的 分析和预测牛带绦虫成虫烯醇酶基因及其编码蛋白的结构和功能,用于指导其生物学功能的实验研究。方法 利用生物信息学网站的各种在线分析工具,并结合其它大型生物信息学分析软件包,如Vector NTI suite,分析从牛带绦虫全长cDNA质粒文库中识别的烯醇酶基因的结构和编码区序列,并分析、预测编码的蛋白质的理化特性、抗原表位、翻译后的修饰、功能域、亚细胞定位、拓扑结构、二级结构、三维空间构象等。结果 该基因全长1 641 bp,编码区为133~1 096 bp,编码320个氨基酸,相对分子量理论预测值为34866.8 Mr。具有一从内到外的跨膜区,无各种亚细胞定位序列,具有多个潜在的磷酸化位点,蛋白的理化性质稳定。有13个主要的B细胞抗原表位。结论 通过生物信息学分析从牛带绦虫成虫cDNA质粒文库中发现烯醇酶基因,并预测得到其结构与功能方面的信息。Objective To analyze and predict the structure and characteristics of enolase by bioinformatics in order to guide the experimental research for its biological function and application..Methods By the bioinformatics analyzing tools in bioinformatics websites and combining else complicated bioinformatics software such as vector NTI suit,so as to recognize enolase from the Taenia solium full-length cDNA plasmid libratory and analyze the gene structure,so as to analyze and predict the physical-chemical characteristics,epitopes,post-translational modification sites,domains,subcelluar location,topological structure,secondary structures,and 3D structural conformation..Results The gene was found to have 1 641 bp,its coding region was 133~1 096 bp,encoding 320 amino acids.The molecular weight of the gene was predicted to be 34866.8 Mr.The protein with stable physical chemical characteristics had a transmembrane region and had no sequence for subcellular location.Several phosphorylation sites and thirteen B-cell eiptopes were predicted in the enolase protein.Conclusions The gene encoding enolase protein was screened form the cDNA library of adult Teania saginata by bioinformatics method.The structure and characteristic Teania saginata of the gene and protein of enolase gene were obtained.
