摘 要:目的观察中药黄芪对早期糖尿病大鼠肾血流动力学的影响及其可能的机制。方法用廓清实验了解黄芪对糖尿病肾病早期肾血流量(RPF)、肾小球滤过率(GFR)的影响;用逆转录半定量聚合酶链式反应(RT-PCR)分析观察黄芪对糖尿病鼠肾诱生型——一氧化氮合成酶(iNOS)基因表达的影响。结果发现4周糖尿病鼠存在RPF、GFR明显升高,用黄芪治疗可使升高的RPF、GFR部分降低;同时发现糖尿病早期肾脏皮、髓质iNOS表达升高,用黄芪治疗则使肾皮、髓质的iNOS表达明显下调。结论黄芪可部分纠正糖尿病早期的肾脏高灌注、高滤过,该作用可能与抑制肾脏一氧化氮(NO)合成有关。Objective To investigate the effects of astragalus membranaceus (Fisch. ) bge. on renal hemodynamicsin early diabetic rats, as well as the possible mechanism involved in. Methods Three groups of rats [ control rats,diabetic rats with and without astragalus membranaceus (Fisch. ) bge. treatment j were followed for 4 weeks. Renalplasma flow (RPF) and glomerular filtration rate (GFR) were examined by clearance study. Inducible nitric oxidesynthase (iNOS) gene expressions in rat kidney were studied by semi--quantitative reverse transcription-polymerase chainreaction (RT-PCR ) analysis. Results Four weeks diabetic rats had supernormal RPF and GFR. Astragalusmembranaceus (Fisch. ) bge. treatment partially lowered increased RPF and GFR. Semiquantitative RT-PCR analysisshowed that early diabetic renal conical and medullary iNOS gene expressions elevated markedly. Renal conical andmedullary iNOS mRNA levels declined obviously in astragalus membranaceus (Fisch. ) bge. treated rats. ConclusionAstragalus membranaceus (Fisch. ) bge. could partially ameliorate diabetic renal hemodynamic abnormalities, in whichinhibition of renal nitric oxide (NO) production might be involved.
