摘 要:目的 观察川芎嗪对内毒素刺激的大鼠肝星状细胞(HSC)增殖和胶原表达的影响,探讨川芎嗪抗肝纤维化的作用机制.方法 于内毒素中刺激大鼠HSC的基础上加川芎嗪分组培养;并采用MTT法检测川芎嗪对HSC生长增殖的抑制作用,免疫细胞化学染色法观察HSC α-平滑肌肌动蛋白(α-SMA)的表达,同时采用ELISA法检测其上清液Ⅰ、Ⅲ型胶原水平.结果 川芎嗪对内毒素刺激的HSC增殖有抑制作用,能下调活化HSC α-SMA的表达,对HSC Ⅰ、Ⅲ型胶原的分泌也有明显抑制作用,并呈剂量依赖性.结论 川芎嗪能抑制内毒素刺激的HSC活化增殖和分泌胶原,为探索川芎嗪抗肝纤维化的分子机制提供了实验依据.Objective To observe the effects of tetramethylpyrazine (TMP) on the proliferation and collagen synthesis of rat HSC stimulated by endotoxin, and to elucidate the anti-fibrotic molecular mechanism of TMP. Methods .Rat HSCs were stimulated by endotoxin and cultured with TMP, then the cultured HSC were divided into four groups. MTT cotorimetric assay was used to detect the proliferation of HSC. The expression of α-SMA was observed by irnmunohistochemical staining.The levels of type Ⅰ and type Ⅲ collagen were detected by enzyme-linked immunosorbent assay (ELISA). Results TMP had a dose-dependent inhibiting effect on the proliferation of HSC stimulated by endotoxin. TMP could down-regulate the protein expression of α-SMA.The levels of type Ⅰ and Ⅲ collagen were also decreased.Conclusion TMP could inhibit the proliferation and collagen synthesis of rat HSC stimulated by endotoxin.it provided experimental evidence for the anti-fibrotic molecular mechanism of TMP.
