嗜酸性乳杆菌细胞壁提取成分对小鼠小肠上皮内淋巴细胞免疫功能的影响

摘　　要：采用机械分离法和Ｐｅｒｃｏｌ非连续密度梯度离心法从鼠小肠提取小肠上皮间淋巴细胞（ｉｎｔｅｓｔｉｎａｌｉｎｔｒａ－ｅｐｉｔｈｅｌｉａｌｌｙｍｐｈｏｃｙｔｅ，ｉＩＥＬ），应用间接免疫荧光染色法进行表面抗原表型测定；同时，应用嗜酸性乳杆菌细胞壁提取物与ｉＩＥＬ细胞共同孵育作用后，检测ｉＩＥＬ细胞的自然杀伤活性和ＩＬ—２产生情况。结果表明：ｉＩＥＬ细胞膜表面抗原的表型７３８％以上为ＣＤ８＋Ｔ细胞，且有４３８％表达ＴＣＲγδ。酸性嗜乳杆菌细胞壁提取物能增加ｉＩＥＬ细胞的自然杀伤活性，并能促进ｉＩＥＬ细胞产生ＩＬ—２。mURINE INTESTINAL INTRAEPITHELIAL LYMPHOCYTE(Iiel)was isolated by using mechanical technique and Percoll discontinuous density gradient centrifugation and its surface antigen waas mesured by using indirect immunofluorescence At the same time,the cell wall of Lactobacillus was cocultured with iIEL and the natural killing activity of iIEL and its ability to produce Interleukin-2(IL-2)were examined Its resultsshowd that more than 73^8%IEL were CD 8 +T cells and more than 43 8%IE were TCRγδ These results confirmed that the cell wall of Lactobacillus could regulate mocosal immunity by enlarging natural killing activity and secreting cytokine as IL-2
