山羊小腔卵泡DNA含量的荧光分析

摘　　要：用显微手术法剥离山羊３ｍｍ以下完整小腔卵泡，用胶原酶和ＴｒｉｔｏｎＸ１００进行处理，选用荧光试剂４，６二脒基２苯吲哚（４，６ｄｉａｍｉｄｉｎｏ２ｐｈｅｎｙｌｉｎｄｏｌ２ＨＣｌ，ＤＡＰＩ），对其ＤＮＡ含量进行分析．结果表明，山羊３ｍｍ以下卵泡的ＤＮＡ含量与卵泡直径相关性极显著（ｒ＝０９８２４＞０７８００＝ｒ００１，ｎ＝１２）．该法可测定５ｎｇ的ＤＮＡ．Intact goat small antral follicles were dissected from ovaries by a micro surgical method. The follicular DNA content was estimated by a fluorometric method using the fluorochrome , 4,6 diamidino 2 phenylindol 2HCl(DAPI).Significant positive correlations were found between the follicular diameters and follicular DNA contents in follicles smaller than 3 mm ( r =0 9824>0 7800= r 0 001 , n =12). It suggested that this method of DNA assay would have great application value in study on follicular gowth and development.
