摘　　要：以全长人ＴＰＯｃＤＮＡ为模板，采用ＰＣＲ方法扩增出ＴＰＯ功能区段基因，克隆入ＰＵＣ１８中，经筛选鉴定后，插入原核表达载体中进行表达，结果ｒｈＴＰＯ功能区片段在原核细胞中获得高效表达，表达量约占菌体总蛋白的４１５％，表达产物经初步纯化后测活，具有明显促进体外培养巨核细胞集落形成的作用。The cDNA of hTPO was extracted as template,functional fragment of hTPO gene was amplified by PCR,inserted into prokaryotic expression vector and transformed into E.coli.The high expression of rhTPO has been achieved.The product of expression acounted for 41.5% of total bacterial protein.After rude purification of the product protein,the biological activity of rhTPO was measured by CFU meg cloning formation test.The results showed that the expressed rhTPO can obviosly stimulate CFU Meg clone formation.The high expression of the activity fragment of hTPO may provide a material to explore its function and clinical usage.
