人胎盘间充质干细胞的分离与鉴定

摘　　要：目的从生产后胎盘中提取胎盘间充质干细胞,并对其进行干细胞特性鉴定。方法采用酶直接消化法从胎盘组织中提取间充质干细胞,并利用流式细胞术及染色体核型分析技术鉴定其干细胞特性。结果成功获得胎盘间充质干细胞,培养48h后即可见贴壁细胞,形态为成纤维细胞样,漩涡状生长,传代过程中细胞形态和特性未见明显变化,经细胞表面分子及染色体核型分析,证明其具备干细胞特性。结论用酶直接消化法所获得的胎盘间充质干细胞具备干细胞特性,可以作为临床干细胞治疗的种子细胞。Objective To explore the Character of Mesenchymal Stem Cells from Human Placenta.Methods The enzyme directory digestion method was used to isolate Mesenchymal Stem Cells from Human Placenta and flow cytometry and karyotyping were used to evaluated the character of stem cells.Results Flow cytometry and karyotyping experiments showed that the cells matched with the character of Mesenchymal Stem Cells.Conclusion The enzyme digestion method can effectively isolate mesenchymal stem cells which could be ideal seed-cells for clinical stem cell implantation.
