摘 要:目的探讨二甲基甲酰胺(DMF)致人肝细胞DNA的损伤情况及不同剂量VitC的干预作用。方法取正常成人肝细胞DMF染毒,DMF剂量为1.56、6.25、25、100 mmol/L、同时设阴性组与H2O2阳性组;100mmol/L DMF染毒后VitC的干预剂量为0.025、0.05、0.10、0.25mmol/L,应用单细胞凝胶电泳技术观察肝细胞DNA的损伤。结果同一处理时间组内各DMF染毒组与阴性对照组间MTL值与MTM值差异均有统计学意义(P<0.05)。不同剂量DMF处理肝细胞后,随着染毒剂量增加MTL值亦增加,最大MTL值出现于100mmol/LDMF组。各染毒组MTL值与MTM值与阴性对照组比较,差异有统计学意义(P<0.05)。结论高浓度DMF能引起肝细胞DNA损伤,小剂量VitC对DMF致肝细胞DNA损伤具有明显的拮抗作用。Objective To explore the DNA Damage of the DMF to liver cell of human being and Intervention of Vitamin C with different doses. Methods Human liver cells were exposed to DMF with 1.56 retool/L, 6.25 mmol/L, 25 mmol/L and 100 retool/L, respectively. Also, a negative group was built and a positive group were exposed to H2O2. Intervetive doses of Vitamin C were 0. 025mmol/L, 0. 05retool/L, 0. 10mmol/L and 0. 25mmol/L after exposure to 100 mmol/L DMF. Single - cell gel electrophoresis was used to observe liver cell DNA damage. Results The MTL value of DNA damage in DMF contaminated groups was significantly higher ( P〈 0. 05) in comparison to the negative group operated in the same time. After exposure to different doses of DMF, the MTL value became larger and larger as contaminated dose increased and the maximum MTL value appeared in 100mmol/L DMF group. In groups with the same dose, except for 25 mmol/L DMF group, the MTM value was significantly higher( P〈 0. 05 ) in the groups of different time, and became larger and larger as working hours increased. Conclusions DMF with high concentration can bring DNA damage to liver cells. Low dose of Vitamin C has obvious antagonism to that effect of liver cell by DMF.
