益气化痰组方对慢性支气管炎模型小鼠的药理学作用研究

摘　　要：目的从药理学角度研究化痰组方对慢性支气管炎模型小鼠抗炎、化痰、平喘、镇咳及机体免疫力的影响,揭示中药作用机制。方法采用烟熏和SO2交替法造模,共计3周。48只小鼠分为模型组、益气化痰方低、中、高剂量组。治疗2周后,观察小鼠咳嗽潜伏期和咳嗽频率的变化;采用爬杆试验观察各组动物运动能力;酚红排痰实验测定痰液的分泌情况。结果造模3周后,与造模前相比,小鼠体重明显降低(P<0.05);小鼠咳嗽潜伏期明显缩短(P<0.05);咳嗽频数明显增加(P<0.05)。治疗2周后,中、高剂量组均能够有效延长小鼠咳嗽潜伏期时间和降低咳嗽频率(P<0.05),但低剂量药物无明显影响(P>0.05)。中、高剂量组均能增加痰液分泌(P<0.01),有明显祛痰作用。各治疗组均能明显延长小鼠爬杆时间(P<0.05)。结论烟熏和SO2交替法烟熏可使小鼠成功患慢性支气管炎。高、中剂量益气化痰组方具有镇咳、平喘、化痰作用;但高、中、低剂量均具备强心、提高机体免疫力和抗疲劳等作用。Objective To investigate on the pharmacological effect of Yiqihuatan prescription on mice with chronic bronchitis and its action mechanism.Methods The mice models with chronic bronchitis were induced by smudging and SO2 alternately for 3 weeks.Then the mice were randomly divided into control group,model group and low,middle and high dose groups.The changes at latent period and cough frequency after 2-week treatment were observed.The climbing pole test was performed to observe the ability of moving after treated,and the secretion amount of phlegm in different groups was detected by phenol red expectoration test.Results The weight of mice was significantly decreased 3 weeks after the models were established,as compared with that before model establishment （P〈0.05）;the latency period of cough was obviously shortened （P〈0.05）; the frequency of cough was significantly increased （P〈0.05）. After 2-week treatment,the middle and high-dose drug groups could effectively prolong the latency period and frequency of cough （P〈0.05）,but the low-dose drug had no obvious effect on them （P〉0.05）.The middle and high-dose drug groups could significantly increase the secretion amount of phlegm （P〈0.01）,and had obvious effect of expelling phlegm. All the groups could obviously prolong the time of climbing pole in the mice （P〈0.05）.Conclusion The smudging and SO2 alternately can induce chronic bronchitis in mice.The synergic effect of different drugs in Yiqihuatan prescription have antiinflammation,antitussive,eliminating sputum,improving the immune ability and antifatigue action.
