摘  要:目的:观察模拟内分泌和外分泌心房钠尿肽(ANP)对胃肠嗜铬(EC)细胞合成与分泌5-羟色胺(5-HT)的影响。方法:通过胃腔直接注射ANP模拟外分泌,通过舌下静脉注射ANP模拟内分泌,采用免疫组织化学染色、透射电镜超微结构的测量学方法和高效液相色谱电化学检测方法观察ANP刺激后胃内5-羟色胺免疫反应细胞密度、内分泌颗粒(SG)数密度及血清5-HT的含量。结果:模拟ANP外分泌,实验组5-HT免疫反应阳性细胞密度、SG数密度明显高于对照组(P<0.05),实验组血清5-HT含量明显低于对照组(P<0.05);模拟ANP内分泌,实验组5-HT免疫反应阳性细胞密度、SG数密度明显高于对照组(P<0.05),实验组血清5-HT含量明显低于对照组(P<0.05)。结论:模拟大鼠内、外分泌ANP可抑制5-HT的分泌,促进5-HT的合成。Objective:To investigate effects of simulating endocrine and exocrine atrial natriuretic peptide (ANP) on synthesis and secretion of 5-HT of enterochromaffin cells (EC) in stomach. Methods:ANP was directly injected into the stomach to simulate exocrine secretion and ANP was injected into the sublingual vein to simulate endocrine secretion. Immunochemical staining, transmission electron microscope and high-performance liquid chromatography-electrochemical detection were used to observe 5-HT expression, dissector of secretory granules (SG) and 5-HT content in serum.Results:Simulating exocrine: compared with control group, the 5-HT expression, the disector of SG were obviously increased in EC cells of rats in experimental group (P〈0.05), but the 5-HT content obviously decreased (P〈0.05). Simulating endocrine: compared with control group, the 5-HT expression, the disector of SG were obviously increased in EC cells of rats in experimental group (P〈0.05), but the 5-HT content obviously decreased (P〈 0.05).Conclusions:Simulating exocrine and endocrine of ANP all can inhibit secretion of 5-HT and promote synthesis of 5-HT.
