芪苈强心胶囊对慢性心力衰竭大鼠PPARα表达的影响

摘　　要：目的观察芪苈强心胶囊对慢性心力衰竭大鼠PPARα表达的影响,探讨其抗心力衰竭作用的相关机制。方法大鼠随机分为假手术组,模型组,芪苈强心胶囊高、中、低组(1.2、0.6、0.3g/kg),每组12只,采用腹主动脉缩窄法制作慢性心力衰竭大鼠模型,术后4周开始灌胃给药,1次/d,连续给药8周。检测血流动力学,处死大鼠,测定血浆中ET-1、Ag-Ⅱ和TNF-α含量,取大鼠心肌组织,RT-PCR和Western blot法检测PPARα mRNA和蛋白的表达。结果芪苈强心胶囊能显著改善血流动力学指标,与模型组比较,芪苈强心胶囊组ET-1、Ag-Ⅱ和TNF-α的含量明显下降(P<0.05或<0.01),PPARα mRNA及蛋白表达均显著上升(P<0.05或<0.01)。结论芪苈强心胶囊可改善心力衰竭大鼠的心功能,与上调PPARα的表达和活性有关。Objective To investigate the effects of Qiliqiangxin capsule on the expression of PPARcx in rats with chronic heart failure, and to explore its molecular mechanisms. Methods The rats were randomly divided into sham operation group, model group, Qiliqiangxin capsule group （ 1. 2,0. 6,0. 3g/kg）, with 12 rats in each group. The rat models with chronic heart failure were induced by transverse abdominal aortic constriction. Four weeks after the constriction, the rats in treatment group were lavaged with Qiliqiangxin for 8 weeks. Hemodynamic parameters and the levels of ET-1 ,Ag- II and TNF-α of plasma were detected. The myocardial tissues were collected and the expression levels of PPARα were detected by RT-PCR and Western Blot. Results Qiliqiangxin capsulecould improve the hemodynamic parameters. As compared with model group, the contents of ET-1, Ag-Ⅱ and TNF-α were decreased, and the expression of PPARoα was significantly increased in Qiliqiangxin capsule group. Conclusion Qiliqiangxin capsule can improve cardiac function in rats by up-regulating the expression of PPARα.
