摘 要:目的观察佐剂性关节炎(adjuvant arthritis,AA)大鼠的一般情况、心功能及神经内分泌免疫指标变化,分析AA大鼠心功能变化与神经内分泌免疫各指标的相关性。方法将24只W ist-ar雄性大鼠随机分为正常对照组和模型组,每组12只,将模型组复制成AA模型。观察两组大鼠一般情况,致炎48 d后通过小动物心功能仪检测心功能,采用ELISA法检测外周血中神经递质:多巴胺(DA),肽类物质:脑钠肽(BNP)、内皮素(ET-1)、降钙素基因相关肽(CGRP),炎症因子:白介素-6(IL-6)、白介素-10(IL-10)、白介素-17(IL-17)和肿瘤坏死因子-α(TNF-α)水平。结果与正常对照组相比,模型组的足跖肿胀度、关节炎指数、H I、LVSP、+dp/dtm ax、HR、TNF-α、DA、ET-1、IL-17、IL-6、BNP、CGRP的表达水平明显升高(P<0.05或P<0.01);体重、-dP/dtm ax、IL-10的表达水平显著降低(P<0.05或P<0.01)。Spearm an相关分析结果显示,AA大鼠心功能各参数变化与神经内分泌免疫各指标有一定的相关性。结论 AA大鼠存在不同程度的心功能降低,并且其心功能变化可能与神经内分泌免疫网络的改变密切相关。Objective To observe the general situation, heart function and the changes in neuroendocrine indexes in the rats with adjuvant arthritis(AA) and analyze the correlation between heart function and every neuroimmunoendocrine index. Methods Divide 24 Wistar male rats randomly into normal control group and model group,12 rats in each one,duplicate AA model in model group;observe the general situation of the rats in two groups, detect heart function with small animal heart function detector 48 days later after inflammation induced, and adopt ELISA assay to detect the levels of neurotransmitters in peripheral blood, including DA, peptides ( BNP, ET - 1, CGRP) and inflammatory factors ( IL - 6, IL - 10, IL - 17 and TNF - α). Results Compared with normal control group, swelling of foot pad, joint inflammatory index, the levels of HI, LVSP, + dp/dt HR, TNF - α, DA, ET - 1, IL - 17, IL - 6, BNP and CGRP all increased apparently in model group( P 〈 0.05 or P 〈 0.01 ) ;the levels of body mass, -dP/dtmax and IL- 10 decreased significantly ( P 〈 0.05 or P 〈 0. 01 ). Spearman correlative analysis indicated that the changes in every parameter of heart function in AA rats were correlative to a certain extent with neuroimmunoendocrine indexes. Conclusion AA rats have declined heart function to different extents. The changes in heart function are probably correlative with the alternations of neuroimmunoendocrine network.
