摘 要:为了探讨大孔吸附树脂纯化鸡血藤中总黄酮的最佳工艺,通过对6种型号大孔树脂的静态实验,筛选出最佳树脂;考察最佳树脂对鸡血藤总黄酮的吸附及洗脱性能,优化工艺参数.结果表明:HZ820为最佳树脂,其纯化总黄酮的优化工艺条件为上样液质量浓度3.31 mg/mL,吸附流速4 BV/h(1 BV为20 mL),上样液体积500 mL,树脂吸附量达79.31 mg/g;以60%乙醇为洗脱剂,洗脱流速3 BV/h,洗脱用量5 BV,解吸率达92.72%,减压浓缩得鸡血藤总黄酮浸膏,纯度为79.49%.To optimize the procedure for total flavonoids purification from Spatholobus suberectus Dunn by macroporous resins,six types of macroporous adsorption resins were compared with their adsorption and desorption properties for total flavonoids.The optimal type of the resin was chosen and the technical parameters for purification of total flavonoids with the chosen resin were studied.Results showed that HZ820 macroporous resin had the best adsorption capacity of 79.31 mg/g under following technological conditions:initial concentration of 3.31 mg/mL,adsorb flow velocity of 4 BV/h(1 BV=20 mL),and the deal capability of 500 mL.The elution rate of 92.72% and the purity of the product of 79.49% could be obtained when the total flavonoids were purged with 60% alcohol of 5 BV on the flowrate of 3 BV/h,the deal capability of 5 BV.
