摘 要:目的建立一种经门静脉胰岛细胞肝内移植治疗远交群(SD)大鼠1型糖尿病的动物模型。方法链尿佐菌素诱导SD大鼠建立1型糖尿病模型20只,分为两组:实验组15只,分别经门静脉输注SD大鼠胰岛1000IEQ(1.5ml);对照组5只,分别经门静脉输注无血清1640液1.5ml。移植术后未服用免疫抑制剂,观察出血量、移植术后血糖、尿量和体重等指标。结果所有大鼠均成活,门静脉穿刺一次成功率90﹪,出血量〈0.5ml。胰岛移植大鼠血糖降为正常的时间为1~6d,平均(3.7±1.7)d。移植胰岛存活时间为2~15d,平均(8.4±4.1)d。移植术后2周内移植组大鼠空腹血清胰岛素明显高于对照组(P〈0.01)。肝脏病理检查显示胰岛细胞在肝窦内存活并能发挥功能。结论大鼠胰岛经门静脉肝内移植是胰岛移植基础研究的理想模型。Objectives To investigate the efficiency of intrahepatic islet transplantation through portal vein in diabetic rats. Methods Type 1 diabetes mellitus was induced with streptozotocin in 20 rats, 15 of which were infused with islets (1000 IEQ, 1.5 ml) through the portal vein respectively as experimental groups and 5 were infused with serum-free 1640 medium (1.5 ml ) respectively as control. No immunosuppressive drug was administered posttransplant. Bleeding volume, blood glucose, urine volume and weight were monitored. Results All rats survived. Success rate of portal puncture was 90 % with bleeding volume less than 0.5 ml. Duration of normal blood glucose posttransplant in experimental group was 1 to 6 days (3.7 ± 1.7 days). The islet graft survival time was 2 to 15 days (8.4 ± 4.1days). Serum insulin levels in experimental group were significantly higher compared with control within 2 weeks posttransplant. Islet graft was viable and functioning in hepatic sinusoids. Conclusions Intrahepatic islet transplantation through the portal vein in rats is feasible.
