摘　　要：目的研究大黄酸(rhein,RH)对白细胞介素6(IL-6)刺激的血管平滑肌细胞(VSMCs)增殖效应及增殖相关因子的影响。方法组织贴块法建立大鼠主动脉原代VSMCs模型并进行鉴定;以MTT比色法观察不同浓度的RH对IL-6诱导的促VSMC增殖效应的干预作用;半定量RT-PCR测定RH对IL-6诱导的VSMCs中PCNA、MMP-3和TIMP1mRNA表达的影响。结果不同浓度RH抑制IL-6刺激的VSMCs增殖效应;RH可抑制IL-6对VSMCs表达PCNAmRNA的上调作用,降低MMP3/TIMP1比值。结论 RH能够抑制IL-6对VSMCs的促增殖作用和表型转换,这可能与调节MMP3/TIMP1比值及PCNA的表达。Objective To observe the effects of rhein on interleukin-6（IL-6）-induced proliferation and correlation factors of vascular smooth muscle cells（VSMCs）.Methods Primary rat aortic vascular smooth muscle cells were cultured with the adherent method of tissue explants and verified.After treated by IL-6 and rhein,the proliferation of VSMCs was determined by MTT assay and the expressions of proliferating cell nuclear antigen（PCNA）,matrix metalloproteinase-3（MMP-3）and tissue inhibitor of metalloproteinase-1（TIMP-1） mRNA were measured by RT-PCR.Results IL-6 significantly promoted VSMCs proliferation in a dose-and time-dependent manner,which was suppressed by rhein.Rhein at different concentrations attenuated the increase of PCNA mRNA and reduced the ratio of MMP-3 and TIMP-1.Conclusion Rhein can inhibit the proliferation and phenotype transition of VSMCs induced by IL-6.The mechanism may be related to the effect of rhein on regulating MMPs/TIMPs and PCNA expression.
