摘 要:目的:观察左归丸和右归丸含药鼠血清对皮质酮所致大鼠海马神经细胞病理模型学习记忆相关信号转导蛋白表达的影响。方法:体外采用高浓度皮质酮(10-4M)处理细胞制作皮质酮损伤型大鼠海马病理细胞模型。采用Westernblot和免疫细胞化学法观察模型细胞与学习记忆相关的信号转导蛋白表达变化及左归丸和右归丸含药鼠血清的干预作用。结果:与空白对照组比较,皮质酮模型组细胞与学习记忆相关的信号转导分子pCaMKII、pCREB、pERK、pCREB和Arc蛋白表达、以及相关受体蛋白GR、MR、TrkB、NR1的表达均明显降低(P<0.05);与皮质酮模型组比较,含10%浓度的左归丸和右归丸鼠血清对模型细胞上述各项指标的异常变化均有显著改善作用(P<0.05)。结论:补肾方药—左归丸和右归丸能够改善高浓度皮质酮所致的海马神经细胞内学习记忆相关信号转导蛋白的异常表达。Objective: To observe the effects of the rat's serum containing Zuogui Wan(左归丸) and Yougui Wan(右归丸) on learning and memory related signal transduction protein repressions in the corticosterone induced rat's hippocampus neural cell pathological model. Methods: The pathological neuron model was established by exposing to the high concentration corticosterone (10-4M) to the primary cultured hippocampus neuron of the newborn 24h SD rat. The learning and memoryrelated signal transduction protein expressions were examined by using Western blotting and immunocytochemistry method, and the effects of Zuogui Wan and Yougui Wan on the model cell were analyzed. Results: Compared with the blank control group, the expressions of pCaMKⅡ, pERK, pCREB signal molecules and Arc protein, and GR, MR TrkB, NR1 receptor proteins were obviously decreased in the pathological neuron model (p〈0.05), there were no significant difference in expression of synapsin-1 of among groups; the rat serum (10%) containing Zuogui Wan and Yougui Wan, however, could improve the abnormal changes of the above indices in the CORT treated cell model obviously (p〈0.05). Conclusion: The Zuogui Wan and Yougui Wan could protect rat's hippocampus neuron from the toxicity of high concentration corticosterone, and ameliorate the abnormal expression of the learning and memory-related signal transduction proteins.
