摘  要:目的:观察慢性阻塞性肺疾病COPD模型大鼠AQPsmRNA表达变化与气道炎症的相关性及"从肠论治"的干预作用,从津液敷布角度探讨COPD"从肠论治"的效应机制。方法:采用气管注脂多糖加熏香烟联合造模方法建立COPD大鼠模型,随机分为正常组、模型组、治肺组、治肠组及肺肠同治组。正常组、模型组灌胃生理盐水,各给药组灌胃相应中药,连续14d。RT-PCR法检测肺组织AQP1、AQP3、AQP5、NF-κBmRNA表达水平。结果:与正常组比较,模型组大鼠气道AQP1、AQP3、AQP5mRNA表达减少(P<0.01),而NF-κBmRNA表达增强;与模型组比较,治肠组AQP1、AQP3、AQP5mRNA表达增强(P<0.05),NF-κBmRNA表达减少(P<0.01);与治肺组比较,肺肠同治组AQP1、AQP3、AQP5mRNA表达增强(P<0.01),NF-κBmRNA表达减少(P<0.01)。气道湿化与炎症程度呈负相关(r=-0.913、-0.304、-0.933,P<0.01)。结论:COPD大鼠水通道AQP5mRNA表达减少,气道湿化功能减弱,气道炎症加重,促使COPD发生发展。而通利大肠,或在治肺的基础上增加通利大肠,可增强AQP5mRNA表达,减少NF-κBmRNA表达,从而改善肺脏津液敷布及宣肺功能,减轻气道炎症,这可能是COPD"从肠论治"效应产生的作用环节之一。Objective:To observe the influence of the mechanism of"treating from intestine"on the expression of AQPs mRNA in rats with COPD,and to investigate the effect of"treating from intestine"from the body fluid metabolic point of view.Methods:The rat model of COPD was established by using the method of cigarette smoking combined with intratracheal injection of LPS.All rats were randomly divided into normal control group,model group,treating lung group,treating intestine group,treating lung and intestine group.The normal control group,model group were given intragastrically the normal saline solution,while other groups were given corresponding herbal drugs intragastrically for 14 days.Detected the expressions of AQP1、 AQP3、AQP5 mRNA in lung by RT-PCR.Results:Compared with normal control group,the expressions of AQP1、AQP3、 AQP5mRNA in airways of model group rats were markedly lower(P0.01),while those of treating lung and intestine group rats were much higher(P0.01).Conclusion:"catharsis large intestine"can enhance the expression of AQPs mRNA in rats with COPD,and therefore improves lung function of fluid metabolic.This may be one of the reasons why"treating from intestine"is effect on COPD.
