栝楼籽核糖体失活蛋白的纯化及性质研究

摘　　要：用改进的方法从栝楼籽制得一种单链核糖体失活蛋白（ＴＣＫ），研究其性质，ＳＤＳ－ＰＡＧＥ和ＩＥＦ显示为单一条带，分子量为２９ｋＤ，ｐＩ≥９．３，含糖量约为１．７５％。在兔网织红细胞裂解液系统中对蛋白质合成有较强的抑制活性，ＩＣ５０为６．７×１０－１０ｍｏｌ／Ｌ。A ribosome inactivating protein, named trichokirin(TCK), isolated and purified to homogeneity from the seeds of Trichosanthes kirilowii by precipitation of (NH4)2SO4, and CMSepharose FF and Sephadex G-50 column chromatography. TCK is a glycoprotein containing 1.75% sugar. The molecular weight of TCK is about 29 kD and pI value is higher than 9.3. Inhibition to protein synthesis was primarily determined by the cell free system of reticulocyte lysate which display the IC50 was 6.710-10mol/L.
