摘 要:目的探讨五味子对去卵巢大鼠骨质疏松症的治疗作用及其机理。方法将59只雌性Wistar大鼠随机分为空白对照组、假手术组、模型组、阳性对照组及五味子组,其中五味子组11只,其余每组12只。摘除大鼠双侧卵巢1个月后,五味子组灌服五味子水煎剂(1g/ml),阳性对照组予己烯雌酚(0.008mg/ml)灌胃3个月(5.6ml/kg)。对不脱钙骨切片进行形态计量并检测胫骨成骨细胞(OB)和骨髓基质细胞(MSC)中护骨素(OPG)、核因子-κB受体活化因子配体(RANKL)蛋白表达。结果五味子组大鼠胫骨骨小梁体积百分比较模型组显著增高,除骨皮质类骨质平均宽度外,骨小梁吸收表面百分比、骨小梁形成表面百分比、骨小梁矿化率术较模型组均显著降低,OB及MSC中OPG蛋白表达明显增高(P<0.05或P<0.01),OB中RANKL蛋白表达明显降低(P<0.05)。结论五味子对卵巢切除大鼠骨质疏松症治疗作用机理之一是通过促进OB和MSC中OPG的分泌,使之与RANKL的结合增多,进而抑制破骨细胞的活性。Objective To explore the effect of Fructus Schisandrae Chinensis on ovariectomy-induced osteoporosis in rats and its functional mechanism.Methods Fifty-nine female Wistar rats were divided into five groups randomly:twelve in the normal control group,twelve in the pseudo-operation group,twelve in the ovariectomy model group,twelve in positive control group,and eleven in the Fructus Schisandrae Chinensis group. One month after bilateral ovariectomy,the rats in the Fructus Schisandrae Chinensis group were administered the decoction of Fructus Schisandrae Chinensis(1g/ml),and those in the positive control group were administered diethylstilbestrol(0.008mg/ml). The treatment lasted 3 months with the dosage of 5.6ml/kg. The bone histomorphometric examination was conducted for the bone histomorphometric analysis with the undecalcified sections made from tibia of rats,and the immunocytochemistry method was applied to detect the expression of osteoprotegerin (OPG) of osteoblast (OB) and marrow stromal cells (MSC) and the receptor activator of nuclear factor-κB ligand (RANKL).Results The percentage of tibial trabecular bone volume in the Fructus Schisandrae Chinensis group was significantly higher than that in the model group. Except for the bony cortex osteoid width,the trabecular bone resorption surface percentage,trabecular bone formation surface percentage,trabecular bone mineralization rate and bony cortex mineralization rate were all significantly lowered than those in the model group; the expression of OPG in OB and MSC was significantly heightened (P〈0.05 or P〈0.01); while the expression of RANKL in OB was significantly lowered (P〈0.05).Conclusion One of the functional mechanisms of Fructus Schisandrae Chinensis in the treatment of ovariectomy-induced osteoporosis in rats is to promote the increase of OPG in OB and in MSC secretion to strengthen their combination with RANKL to inhibite the activity of osteoclast.
