摘  要:目的:应用纤维支气管镜将结核分枝杆菌(MTB)接种猕猴肺部,建立结核病模型。方法:分别用不同菌量的MTBH37Rv株,Erdman株和临床耐药株、经纤维支气管镜接种实验猴右肺下叶,在规定的时间,观察其结核菌素皮肤试验和胸部X线表现、肺部病理学改变和肺组织细菌载量。结果:所有感染猕猴的结核菌素皮肤试验呈阳性,胸部X线呈现结核样改变,病理解剖显示肺部均有不同程度的病理变化;显微镜下肺组织中可见特征性的结核结节,肺组织中均培养出了数量不等的结核分枝杆菌。结论:经纤维支气管镜将一定量的结核分枝杆菌接种实验猕猴的右肺能成功建立结核病实验猴模型,该方法能为结核疫苗和药物疗效评价平台提供良好的技术支撑。Objective:To establish nonhuman primate model for mycobacterium tuberculosis(M.tb)infection by inoculating macaque with M.tb using a fiberoptic bronchoscope to deliver the inoculum into the lung.Methods:Macaques were infected respectively with different doses of M.tb H37Rv strain,M.tb Erdman train,and M.tbclinical drug resistance strain using a bronchoscope to deliver the inoculums directly into the right caudal lung lobes of animals.At regular intervals,skin tests with OT were performed and chest X-ray photos were taken.Histopathologic changes on lung tissues were evaluated.Bacterial loads were also determined on lung lobes.Results:The results of tuberculin skin tests for all of the infected animals were positive.The chest X-ray exhibited pneumonia with nodularity.At necropsy,the animals showed tuberculous pathologic changes on the lungs with different degree.Characteristic tubercles were seen in lung tissues under microscope.Bacterial loads were determined respectively with different colony-forming units(CFU)counts on lung lobes of these infected animals.Conclusion:Nonhuman primate model for M.tbinfection can be established successfully by infecting macaques with M.tb using a bronchoscope to deliver the inoculum directly into the right lung of macaque.This method can provide a good technical support to the platform for evaluating efficacy of vaccines and drugs against tuberculosis.
