摘 要:目的探讨醋酸铅对未成年雄性大鼠骨髓间充质干细胞生长的影响,阐明铅对体内生理情况下骨髓的毒作用机制。方法分别用15、30和60 mg/kg浓度组的醋酸铅灌胃染毒未成年雄鼠,饲养8周。取股动脉血测其铅浓度,处死取双侧股骨骨髓密度梯度离心法体外培养。观察铅对骨髓间充质干细胞成集落能力(CFU-F)和细胞形态、生长的影响及其变化规律。结果 各实验组与对照组比较,血铅浓度的差异均有统计学意义(P<0.05),且随醋酸铅浓度的增加而升高,呈剂量-效应关系(r=0.983,P<0.01)。骨髓间充质干细胞集落形成率(CFU-F)的各组间差异有统计学意义(P<0.05),且随铅的浓度增加抑制作用增强,呈剂量-效应关系(r=-0.967,P<0.01),其IC50≈60.00μmol/L。MTT结果:高剂量染铅组增殖速度要比对照组慢(P<0.05)。结论在本试验条件下,醋酸铅可明显抑制雄性大鼠骨髓间充质干细胞(BMSC)的生长和形态长,并随剂量的增大抑制作用明显增强。Objective To explore the effect of lead acetate on the growth of immature male rats bone marrow mesenchymal stem cells and Clarify the toxicity mechanism of lead acting on bone marrow in physiology circumstance.Method Different doses lead acetate(15、30、60 mg/kg) were fed to immature male rats exposed orally.8 weeks later,The lead levels of femoral artery blood were measured.Rats were killed and bone marrow of the bilateral femur was cultured by density gradient centrifugation in vitro.The effects of lead acetate on the rate of colony formation(CFU-F)、cell morphology and cell growth of bone marrow mesenchymal stem cells and the rule of variation were observed.Result Comparing diferent doses groups with the control group,there were significant differences of blood lead(P 0.05) and dose-effect relationship demonstrates negative correlation(r = 0.983,P 0.01).With the increase of lead concentration the rate of colony formation(CFU-F) decreased in a dose-dependent relationship(r =-0.967,P = 0.000).Meanwhile,the differences of between diferent dosesgroups with the control group were remarkable(P 0.05).The IC50 of CFU-F colony was 60.00 μmol/L.The results of MTT revealed that the proliferation velocity of high-dose lead group was slower than that in the control group(P 0.05).Conclusion The data suggests the growth and morphology of BMSC were significantly inhibited by lead acetate,further,the inhibitory effect enhances with the increasing of lead acetate doses.
