摘 要:目的:探讨二磷酸腺苷(ADP)对细胞的促生长作用。方法:用间接免疫荧光法观察ADP对BSC-1细胞骨架成分的影响。结果:在用ADP前,荧光下非洲绿猴肾上皮细胞(BSC-1)的细胞骨架中角质蛋白纤维成分8(Cytokeratin8,CK8)以星形位于细胞核周围,ADP作用2min后,CK8开始伸长,6h后伸长到最大程度,充满整个细胞,12h后开始萎缩,24h后CK8又以星形位于细胞核的周围。而其它细胞骨架成分未见变化。结论:ADP是肾小管上皮细胞最强的一种有丝分裂源。Objective: Our aim was to evaluate the role of adenosine diphosphate (ADP) on the cytoskeleton of kidney epithelial cells. HZMethods: We used indirect immunoflurescence method to observe the influence of ADP on the cytoskeleton of BSC1 cells. Results: In untreated confluent cells, cytokeratin 8 (CK8) was predominantly confined to starlike spot in the area around the nucleus, but the spot began to spread within 2 minutes after addition of ADP. This process completed at the 6th hour after addition of ADP and a welldeveloped filamentous network throughout the cell formed. At the 12th hour, the network began to collapse and the filaments formed a spot again in the perinuclear area. The process completed by the 24th hour. We did not find any change of other cytoskeleton. Conclusion: Exogenous adenosine diphosphate (ADP) was the most potent growth mitogen for nontransformed kidney epithelial cells of BSC1 line.
