麻杏石甘汤对A型流感病毒感染的MDCK细胞活性与超微结构的影响

摘　　要：目的研究麻杏石甘汤对A型流感病毒感染的MDCK细胞活性与超微结构的影响。方法 (1)运用细胞培养技术和MTT法检测麻杏石甘汤预防给药和治疗给药方式下的流感病毒感染的MDCK细胞活性;(2)用透射电子显微镜观察最大无毒限毒的麻杏石甘汤治疗给药方式下的流感病毒感染的MDCK细胞的超微结构。结果治疗给药方式中,麻杏石甘汤在浓度为31.25 mg/mL时,MDCK细胞活性的OD值显著高于病毒对照组,差异有统计学意义(P<0.05),TI为2.78;预防给药方式中,麻杏石甘汤在浓度为15.63、31.25 mg/mL时,MDCK细胞OD值高于病毒对照组,差异具有统计学意义(P<0.01),TI为1.71;与病毒对照组比较,麻杏石甘汤治疗给药方式下的MDCK细胞超微结构较完整。结论麻杏石甘汤在预防给药和治疗给药方式中均能发挥抗流感病毒作用;并能通过抑制病毒增殖的方式而保护流感病毒感染的MDCK细胞活性与超微结构。Objective To study the effects of Maxing Shigan decoction（MXSGD） in vitro on activity and ultrastructure of MDCK which infected by influenza virus A.Methods ①The activity and ultrastructure of MDCK infected by influenza virus A prevented and treated with MXSGD were determined with the cell culture technique and MTT test.②The ultrastructural of MDCK in that same condition was observed with transmission electron microscopy.Results In the treatment condition,when MXSGD in the concentration of 31.25mg/mL,the OD values of MDCK were significantly higher than that in the virus control group（P0.05）,TI is 2.78;In prevention condition,when MXSGD in the concentration of 15.63mg/mL and 31.25 mg/mL,the OD values of MDCK cells were significantly higher than thaht in the virus control group（P0.01）,TI is 1.71.Compared with the virus control group,the ultrastructure of MDCK cell in treatment condition with MXSGD was more complete.Conclusion MXSGD used for prevention and treatment is able to play the role of anti-influenza virus and inhibit the proliferation of the virus to protect cell activity and ultrastructure of MDCK which infected by influenza virus A.
