摘 要:目的研究甘草黄酮(LF)对高脂高糖饮食结合一次性小剂量腹腔注射链脲佐菌素(streptozotocin,STZ)所致糖尿病大鼠高血糖与高血脂的预防作用,探讨其可能的作用机制。方法将实验大鼠随机分成4组:正常对照组(control)、正常给药组(CLF)、模型组(model)和预防给药组(PLF)。control组和CLF组以普通饲料饲养,model与PLF组以高脂高糖饲料饲养;同时,CLF组与PLF组以剂量为300mg(/kgbody.d)的LF灌胃,control组与model组以溶剂(1,2-丙二醇)灌胃。六周后,model组和PLF组一次性小剂量腹腔注射STZ(35mg/kg)。6天后检测实验大鼠体重、空腹血糖(FBG),甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)和总抗氧化能力(T-AOC);取肝脏、肾脏、肾周脂肪、双侧附睾脂肪和大网膜脂肪称重;计算大鼠的肝指数、肾指数及脂体比。结果LF显著抑制了PLF组大鼠的FBG、TG、TC、LDL-C和肝体比的升高,显著提高了T-AOC,并显著抑制了PLF组大鼠体重和脂体比的下降。结论LF可有效预防大鼠实验性糖尿病的发生。【Objective】 To study the preventive effect of licorice flavonoids (LF) on hyperglycemia and hyperlipemia of experimental diabetic rats induced by feeding high-fat and high-sugar diet combined with low-dose streptozotocin (STZ) injection,and to elucidate its possible pharmacological mechanisms.【Methods】The rats were divided into four groups,namely as control group,control treatment group (CLF),model group and preventive treatment group (PLF). Rats in control group and CLF group were fed with normal diet,while rats in the other two groups were fed with high-fat and high-sugar diet. Meanwhile,rats in CLF and PLF groups were administered with 300 mg(/kg body·d)of LF. Rats in control and model groups were administered with solvent (1.2-propanediol). 6w later,the model group and PLF group were injected STZ (35 mg/Kg). 6d later,the body weight,levels of the fasting blood glucose (FBG),triglycerides (TG),total cholesterol (TC),high-density lipoprotein cholesterol (HDL-C),low-density lipoprotein cholesterol (LDL-C) and total antioxidation capability (T-AOC) of all experimental animals were examined. Liver,kidney,fat around kidney,epididymis and epiploon were weighed. Liver index,kidney index and the relative weight of abdominal adipose tissue were calculated. 【Results】LF preventive treatment significantly inhibited the increase in levels of FBG,TG,TC,LDL-C and liver index,it also significantly increased the levels of T-AOC and inhibited the decrease in body weight and relative weight of abdominal adipose tissue.【Conclusion】 LF is effective in preventing occurrence of experimental diabetes in rats.
