摘 要:目的观察糖尿病周围血管病患者内皮祖细胞(endothelial progentior cells,EPC)一氧化氮(NO)、内皮型一氧化氮合酶(eNOS)表达的变化。方法采用硝酸还原酶法测定NO的浓度,免疫印迹杂交法(Westernblot)半定量测定eNOS的含量。结果糖尿病合并周围血管病组EPCs数量最少,增殖、迁移和黏附能力降低,NO和eNOS表达最弱,糖尿病无血管病组表达增强,对照组表达最多,差异具有统计学意义(P<0.05)。NO和eNOS的表达与EPC的数量呈正相关(r=0.651,P<0.05)。结论糖尿病合并周围血管病患者EPC数量减少,功能降低,可能与eNOS、NO表达减弱有关。【Objective】To investigate expressions of NO and eNOS in endothelial progenitor cells(EPC) of type 2 diabetes mellitus patients(T2DM) with peripheral angiopathy.【Methods】NO(nitric oxide) secretion of EPCs was measured by Griess assay with spectrophotometer.The concentration of endothelial nitric oxide synthase(eNOS) was assayed by Western blot.【Results】 In T2DM group with peripheral angiopathy,the amount of EPCs was the smallest and the capacity of proliferation,adhesion and migration were significantly decreased in the group of T2DM with peripheral angiopathy;and the expressions of NO and eNOS were greatly suppressed in the T2DM group with peripheral angiopathy,enhanced in the T2DM group without peripheral angiopathy and the most prominent in the control group with significant difference(P0.05).The amount of EPCs showed a positive relationship with the expressions of NO and eNOS(r=0.651,P0.05).【Conclusion】The mechanism of diabetic peripheral angiopathy may be correlated with the attenuation of the expressions of NO and eNOS.
