摘 要:目的研究维吾尔药神香草对哮喘大鼠血清白介素-17(IL-17)水平及Th1/Th2平衡的影响,阐明神香草治疗哮喘的免疫机理。方法将大鼠随机分为正常对照组、哮喘模型组、地塞米松治疗组及神香草水提液高、低剂量治疗组。采用卵清白蛋白(OVA)﹑氢氧化铝[Al(OH)3]及百白破疫苗联合致敏和OVA生理盐水雾化激发的方法制备哮喘模型。采用酶联免疫吸附试验(ELISA)法检测大鼠血清中IL-17、白介素-4(IL-4)及干扰素-γ(IFN-γ)的水平。结果正常对照组与哮喘模型组、哮喘模型组与治疗组血清IL-17﹑IL-4及IFN-γ的水平差异均有显著性(P<0.05);神香草高剂量治疗组降低细胞分泌IL-17的作用大于低剂量治疗组(P<0.05);与模型组比较,神香草治疗后哮喘大鼠血清IL-4的水平显著降低(P<0.05),而IFN-γ的水平显著增高(P<0.05)。各组大鼠血清IL-4与IFN-γ水平呈密切负相关(r=-0.997),IL-17水平与IL-4和IFN-γ水平呈负相关关系(r=-0.333,-0.509),均P<0.05。结论神香草可通过抑制IL-17的分泌,纠正Th1/Th2型细胞因子失衡,发挥抗炎作用。【Objective】To study the Uygur medicine Hyssopus officinalis L on serum IL-17,IL-4 and IFN-γ levels of asthma rats in order to explore the mechanism of its treatment of asthma【.Methods】 Rats were randomly divided into normal control group,asthma model group and dexamethasone group and water extract of Hyssopus of-ficinalis L high and low dose group.The rats were sensitized with OVA,AL(OH) 3 and DPT vaccine and then challenged with inhalation of aerosolized OVA solution for Preparation of asthma model and the level of serum IL-17,IL-4 and IFN-γ were detected with ELISA.【Results】 The normal control group and model group,model group and treatment group,the secretion of IL-17 levels were statistically significant differences(P〈0.05);Hyssopus officinalis L high-dose treatment group reduce cell secretion of IL-17 was stronger than the low-dose treatment group(P〈0.05).Compared with model group,after treatment of Hyssopus officinalis L serum IL-4 levels of asthma rats significantly reduced(P〈0.05),but IFN-γ level was significantly higher(P〈0.05).The serum IL-4 and IFN-γ levels are closely negative related(r=-0.997),the serum IL-17 level has negative correlation with IL-4 and IFN-γ levels(r=-0.333,-0.509).【Conclusion】Hyssopus officinalis L can inhibit the secretion of IL-17 and correct the imbalance between Th1/Th2 cytokines to play the role of anti-inflammatory.
