摘 要:目的观察己酮可可碱对大鼠脊髓和背根神经节(DRG)肿瘤坏死因子-α(TNF-α)的影响。方法雄性SD大鼠40只,体重180~220g,分为7组。对照组(10只)经口灌胃生理盐水(NS)2mL/次,每天1次;A、B、C组(各5只)按17mg/kg;D、E、F组(各5只)按8.5mg/kg经口灌胃HgCl2溶液,每天1次。7组均连续灌胃18~22d。染汞组动物出现汞中毒症状后,处死A、D组大鼠和对照组5只大鼠。随后B、E组用二巯丙磺钠注射液(DMPS)按28mg/kg腹腔注射驱汞2个疗程(驱汞3d,休息4d为1个疗程);C组用DMPS+己酮可可碱(按29.22mg/kg经口灌胃,每天1次,共14d);F组腹腔注射NS1mL/次,方法同DMPS。实验结束后处死全部动物,灌注固定取出脊髓L5-6节及L5、L6两侧DRG并制成病理切片。用免疫组织化学SABC法测定DRG和脊髓TNF-α表达水平。结果染毒组大鼠DRG和脊髓TNF-α平均灰度均显著低于对照组;阳性反应物平均面积均显著高于对照组(P<0.05);B、E组用DMPS驱汞2个疗程及F组腹腔注射NS2周期后,均未见TNF-α表达显著减弱,C组(DMPS+己酮可可碱)DRG和脊髓TNF-α平均灰度显著提高,平均面积减少(P<0.05)。结论亚急性HgCl2中毒大鼠DRG和脊髓TNF-α表达显著增强,己酮可可碱联合DMPS能够有效抑制TNF-α表达。【Objective】To investigate the effect of pentoxifylline on tumor necrosis factor-α (TNF-α) in spinal cord and dorsal root ganglion (DRG). 【Methods】Forty SD rats (male, weighting 180-220 g) were divided into 7 groups. Group A, B and C (5 rats in each group) received HgCl2 juice at dose of 17 mg/kg (1/4 LD50), group D, E and F (5 rats in each group) received HgCl2 juice at dose of 8.5 mg/kg (1/8 LD50) by gavage daily. Control group (10 rats) received normal saline (NS) 2 mL by gavage daily. The sub-acute mercury poisoning rat models were established within (20±2) days. Then five rats in each of control group, group A and group D were dissected and the chelator (DMPS) was injected into the abdomen of the alive rats of group B and E at dose of 28 mg/kg for 2 periods. Group C was given both DMPS and pentoxifylline and group F was injected 1 mL NS into the abdomen of rats only. The pentoxifylline was used at dose of 29.22 mg/kg by gavage daily for 14 days. The rats were all dissected and the spinal cord (L5-6) and the DRG of L5 and L6 were gotten after the trial was finished, and the pathological sections were made. TNF-α was detected according to immunohistochemistry SABC way. 【Results】The mean gray scale value of TNF-αin spinal cord and DRG of the rats exposed to HgCl2 was decreased significantly compared with that of control group (P 0.05). They are not decreased significantly after two 7-day cycles for group F using NS and groups B and E using DMPS compared with control group. The mean gray scale value of TNF-α in spinal cord and DRG of group C was increased significantly after 2 periods of DMPS and pentoxifylline administration compaired with that of group B (P 0.05).【Conclusion】 TNFαlevels in spinal cord and DRG of sub-acute mercurialism rats are increased significantly compaired with that of controls and they are inhibited significantly by pentoxifylline combined with DMPS.
