摘  要:目的探讨脂联素基因多态性与2型糖尿病的相关性。方法本研究入选昆明地区汉族2型糖尿病患者133名,否认糖尿病家族史的非糖尿病个体57名。采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法检测脂联素基因SNP-11377、SNP+45和SNP+276基因型,构建单倍型。评估以上三个多态性位点与2型糖尿病的相关性。结果①脂联素基因SNP-11377、SNP+45和SNP+276的基因型和等位基因频率在昆明地区汉族2型糖尿病组和非2型糖尿病组中无统计学差异。②SNP-11377、SNP+45和SNP+276所构建的单倍型频率在昆明地区汉族2型糖尿病组和非2型糖尿病组中无统计学差异。③在非2型糖尿病组中,SNP-11377CC基因型个体的体重指数和腰围较低。结论脂联素基因SNP-11377、SNP+45和SNP+276与昆明地区汉族2型糖尿病无相关性。SNP-11377CC基因型可能是昆明地区汉族非糖尿病个体肥胖的一个保护基因型。【Objective】To investigate the correlation between adiponectin gene polymorphisms and type 2 diabetes(T2DM).【Methods】Totally 133 patients with type 2 diabetes and 57 nondiabetes(NDM) subjects with no family history of diabetes in Kunming,Yunnan Province,Southwest of China,were included in the present study.Using polymerase chain reaction-based restriction fragment length polymorphism(PCR-RFLP),the genotypes of SNP-11377,SNP+45 and SNP+276 in adiponectin gene were detected and their haplotypes were constructed.Then,the correlation between the three SNPs and T2DM of Han population was assessed.【Results】 The genotype and allele frequencies of SNP-11377,SNP +45 and SNP +276 had no significant difference between T2DM and NDM groups.The constructed haplotype frequencies of SNP-11377,SNP+45 and SNP+276 had no significant difference between T2DM and NDM groups.In NDM group,subjects with SNP-11377CC genotype had lower body mass index(BMI) and waist circumference.【Conclusions】There is no correlation between the SNP-11377,SNP+45,SNP+276 and T2DM of Han population in Kunming.SNP-11377CC genotype might be a protective genotype against obesity in NDM group of Han population in Kunming.
