摘  要:目的探讨大肠杆菌(E.coli)感染与人巨噬细胞系U937细胞凋亡的关系及热休克蛋白70(HSP70)、核因子(nuclear factor kappa B,NF-κB)表达的变化。方法以Annexin V FITC/PI双染流式细胞仪检测为指标,研究E.coli感染对人巨噬细胞系U937细胞凋亡的诱导作用;用Western blot方法检测HSP70和NF-κB的表达。结果Annexin V FITC/PI双染结果发现,U937细胞凋亡百分率随E.coli浓度的增加而增加,HSP70的表达随着E.coli浓度的增加逐渐升高,而NF-κB的表达逐渐降低。结论E.coli诱导U937细胞凋亡过程可能与HSP70表达的升高和NF-κB表达的降低有关。【Objective】To investigate the relationship between apoptosis of human monocytic U937 cells and their expression of heat shock protein 70 kd(HSP70) and nuclear factor kappa B(NF-κB) induced by Escherichia coli(E.coli).【Methods】Cell apoptosis was analyzed by Annexin V FITC/PI assay.Western blot was used to detect the expressions of HSP70 and NF-κB protein.【Results】Annexin V FITC/PI assay revealed that the percentage of apoptotic cells rose with the increasing concentrations of E.coli.Also,there was a tendency for HSP70 protein expression to increase and for NF-κB protein expression to decrease with the increased E.coli concentrations.【Conclusions】U937 cell apoptosis induced by E.coli may be related to the increase of HSP70 expression and decrease of NF-κB expression.
