摘  要:由于光捕获天线蛋白(LHCⅡ)编码基因(lhcb)家族同源性较高,以往研究只能从总体上反映家族基因的变化,对各个编码基因的表达研究较少.本研究以光合单细胞生物盐生杜氏藻(Dunaliella salina)为材料,应用实时PCR的方法,系统研究了lhcb基因家族成员在光环境转换过程中的表达变化.结果表明,当由正常培养光环境转移到高光条件下时,lhcb基因家族成员的表达整体上受抑制,但各个基因的表达变化趋势随时间变化不尽相同.在强光照下,lhcb1、lhcb2.1、lhcb2.23h时有最低的表达积累量,而lhcb3却有较高的表达量.细胞转入黑暗环境后,各个基因的转录水平升高,变化趋势基本相同,在3h时均有较高的表达量,只有lhcb2.1在1h时的表达量最高.叶绿素加氧酶编码基因cao的变化与lhcb3基本相同.LHCⅡ蛋白量在高光下降低,在黑暗条件下升高.叶绿素含量在高光条件下明显下降,叶绿素a/b值升高,黑暗条件下则相反.Due to the high homology among the family members encoding light-harvesting protein(lhcb),only total gene expressions,not the individual gene of lhcb were studied in previous researches.Using the real-time PCR,this study investigated the gene expression changes of the lhcb members under different light intensities in single-cell photosynthetic organism Dunaliella salina.The results showed that the expressions of lhcb were inhibited after the transition to high light intensity of cells,but there was no similar changing tendency for each gene with the change in time.Contrary to the lowest accumulation of lhcb1,lhcb2.1 and lhcb2.2 in 3 h,the mRNA amount of lhcb3 was the highest except the control(0 h).When cells were transferred to the dark condition,the mRNA amount of each gene elevated and existed a similar changing current,which peaked in 3 h except lhcb2.1 in 1 h.The expression change of cao encoding chlorophyll a oxygenase was similar to that of lhcb3.The accumulations of LHCⅡ and chlorophyll decreased and elevated under high light intensity and dark condition,respectively,but the value of chla/b showed a contrary changing tendency.
