摘  要:目的:观察补髓生血颗粒对CAA患者骨髓单个核细胞中FAK表达及FAK磷酸化水平变化,探讨补髓生血颗粒治疗CAA的作用机制。方法:将60例CAA患者予以补髓生血颗粒治疗6个月,另取20例健康志愿者作为正常对照组。采用蛋白质印迹方法(Western-blot)观察CAA患者治疗前后骨髓单个核细胞中FAK及FAK pY397的表达水平。结果:CAA患者治疗前骨髓单个核细胞FAK及FAKpY397的表达均低于正常对照组(P均<0.05),经补髓生血颗粒治疗后,FAK及FAK pY397的表达均所上升,与治疗前比较均具有显著性差异(P均<0.05)。结论:CAA患者骨髓单个核细胞FAK及FAKpY397的水平均呈低水平表达,提示FAK与CAA发病密切相关,补髓生血颗粒可能通过调节FAK及FAK pY397的表达来改善CAA骨髓造血微环境。Objective: To observe the effect of Busuishengxue Granule on the expression of FAK and the level of Phosphorylated FAK in CAA patients' bone marrow,and to explore the mechanism of Busuishengxue Granule on CAA.Methods: 60 CAA patients were treated with Busuishengxue Granule for 6 months,and another 20 healthy volunteers were as normal control group.Use Western blot method(Western-blot) to observe the expression of FAK and FAKpY397 in CAA patients' bone marrow mononuclear cells before and after treatment.Results: The expression of FAK and FAK pY397 in CAA patients'bone marrow mononuclear cells were lower than the normal control group(P all 0.05),and expression of FAK and FAK pY397 were both on the rise after treatment,so they took on significant differences compared with them before treatment.Conclusion: The expression of FAK and FAK pY397 in CAA patients'bone marrow showed low levels,so it suggested that FAK was closely related to the pathogenesis of CAA,and Busuishengxue Granule may regulate the expression of FAK and FAK pY397 to improve the CAA bone marrow microenvironment.
