针药并用对单纯性肥胖大鼠瘦素含量及瘦素受体表达的影响

摘　　要：目的:观察中药石连饮+针灸对肥胖大鼠血清及下丘脑瘦素含量和下丘脑瘦素受体表达的影响,探讨针药并用治疗单纯性肥胖的机制。方法:Wistar雄性大鼠120只,随机分为普食组及高脂组,高脂组用高脂饲料喂养16w制作肥胖大鼠模型。将造模成功的60只大鼠随机分为:石连饮+针灸组、西药组、模型组。针刺大鼠"后三里"和"中脘"穴,并接通电针仪,并灌喂中药石连饮15g.kg-1,每日1次,连续治疗14d。每10d测体重、体长并计算Lee's指数。放射免疫法检测血清瘦素含量,免疫组织化学法检测下丘脑瘦素受体表达。结果:与模型组比较,针药组血清瘦素含量降低(P<0.01),下丘脑瘦素受体表达增加(P<0.01)。结论:针药并用对肥胖机体下丘脑和血清中leptin良性调节作用是改善瘦素抵抗以及调节异常内分泌代谢的作用机制之一。Objective： To observe the effect of Acupuncture combined with traditional Chinese medicine on serum and Hypothalamus leptin contents and the expression of leptin receptor in the hypothalamus in simple obesity rats so as to explore the underlying mechanism of Acupuncture combined with traditional Chinese medicine in treating Simple Obesity Rats.Methods： A total of 120 male Wistar rats were randomly divided into normal control,high grease groups.The high high grease group was established by feeding 16w the animal with high fat forage.The 60 rats which were made model blest were randomly divided into shilianyin ＋ electro-acupuncture groups、western medicine groups、model groups.electro-acupuncture was applied to ＂Zusanli＂and ＂Zhongwan＂,and pour Shilianyin15g.Kg-1,once daily,for14 days.The body weight,body length and Lee＇s index were measured once every 10 days.Serum leptin contents were measured by radioimmunoassay,and leptin receptor expression was detected with immunohistochemistry.Results： In comparison with normal control group,the serum leptin content of model group decreased significantly（P 0.01）,the number of leptin receptor expression increased considerably（P 0.01）.Conclusion： The optimum accommodative role of acupuncture combined with traditional Chinese medicine to Hypothalamus and Serum leptin of obesity body is one of the mechanism of action to improve leptin-counteract and accommodate sabnormal internal hormone metabolism.
