摘 要:以细叶百合的鳞片和花丝为外植体,通过两种不同的发生方式,成功获得再生植株,并建立了细叶百合两种外植体的再生体系。结果表明:鳞片可以通过器官直接发生途径再生,其最适诱导培养基为MS+NAA0.5mg·L-1+6-BA1.5mg·L-1;花丝通过愈伤组织间接器官发生途径再生,诱导愈伤组织的最适培养基为MS+2,4-D2mg·L-1+KT2mg·L-1,诱导愈伤组织产生不定芽的最适培养基为MS+NAA0.5mg·L-1+6-BA2.0mg·L-1;最适继代增殖培养基均为MS+NAA0.2mg·L-1+6-BA1.0mg·L-1;最适生根培养基为1/2MS+IBA0.5mg·L-1。Regeneration plants were induced successfully in plants of Lilium pumilum using bulb scale and filament as explants. An efficient regeneration system was developed through organogenesis in the two kinds of explants. Results showed that shoots were induced from bulb scale directly,and the optimal medium for bulb scale induction was MS medium supplemented with 0.5mg/L NAA and 1.5mg/L 6-BA. However,callus was obtained from filament explants via organogenesis indirectly,and MS medium supplemented with 2mg/L 2,4-D and 2mg/L KT was the optimal medium for callus induction,but the highest regeneration rate of shoots was achieved on MS medium containing 0.5mg/L NAA and 2.0mg/L 6-BA. For both bulb scale and filament induction,MS medium supplemented with 0.2mg/L NAA and 1.0mg/L 6-BA was suitable for subculture,and 1/2MS with 0.5mg/L IBA was optimal for rooting.
