摘 要:对散斑壳属2个近似种,即松针散斑壳(Lophodermium pinastri)和乔松散斑壳(L.pini-excelsae)的外部形态及内部构造进行了比较研究,并绘图示之。同时对此二种多个菌株的rDNA-ITS序列进行了PCR扩增与测定,并与GenBank中已有的相关序列进行同源性分析;利用MEGA邻接法(neighbour-joining method,NJ)构建了系统发育树。结果表明,在形态解剖学方面,L.pinastri除子座基部层、线纹及分生孢子器开口特征外,其余重要特征与L.pini-excelsae具有很大的相似性。L.pinastri ITS序列的G+C含量(51.5%)大于L.pini-excelsae的G+C含量(50.1%)。从系统发育树可以看出,双维松上的L.pinastri和单维松上的L.pini-excelsae形成2个明显独立的分支,寄主对种内遗传多样性也产生较重要影响,产地与种内遗传差异无明显关联。ITS序列分析可以作为区分这2个近似种的一种有效手段。Macroscopic appearance and internal structure of two related species of Lophodermium pinastri and L. pini-excelsae were described and illustrated. At the same time,rDNA-ITS sequences of some strains of the two species were amplified and determined,and the genetic homology among these sequences and other related sequences in GenBank was analyzed. A phylogenetic tree of these strains was constructed by MEGA using neighbour-joining method. Results show that the internal characteristics of L. pinastri are remarkably similar to those of L. pini-excelsae,except those of stroma basic layer,zone lines and opening of pycnidia. G+C content of L. pinastri(51.5%) was higher than that of L. pini-excelsae(50.1%). The phylogenetic tree indicates that L. pinastri on diploxylon pines and L. pini-excelsae on haploxylon pines can be sharply classified into two independent groups,and hosts have a definite impact on intraspecific genetic diversity. Origin is not significantly associated with intraspecific genetic variation. ITS sequence analysis is an effective means to distinguish the two related species.
