摘 要:酶联免疫吸附试验(ELISA)是一种常用的固相酶免疫测定方法,但ELISA的影响因素较多,必须加强质量控制才能充分发挥其方法学的优点。为了检测ELISA方法在检测氯霉素残留中的应用效果,从精密度、灵敏度、特异性、板内差异多个角度对酶联免疫测定法进行质量评价。结果表明:灵敏度可达到小于0.025 ng/mL,检测范围为0.025~100 ng/mL,板内变异系数CV<20%,与结构相似的甲砜霉素、氟甲砜霉素没有交叉反应。The enzyme-linked immunosorbent assay(ELISA) is a commonly used method of immobilized enzyme immunoassay.There are many factors associated with ELISA,quality control must be strengthened to fully play the methodological advantages of ELISA.In order to test the application effects of ELISA in the detection of chloramphenicol residues,the authors evaluated the performance of ELISA in terms of precision,sensitivity,specificity and intraplate difference.The results showed that the sensitivity was less than 0.025ng/ml,the detection range was 0.025-100 ng/mL,the intraplate coefficient of variation was less than 20% and chloramphenicol did not have cross reaction with thiamphenicol and florfenicol,which was similar to chloramphenicol in structure.
