摘  要:目的探讨郁金抗四氯化碳所致急性肝损伤小鼠肝细胞凋亡机制。方法 ICR小鼠48只,雌雄各半,随机均分6组:空白对照组(生理盐水),模型组(生理盐水),阳性药组(甘利欣22.5 mg.kg-1),郁金高、中、低剂量组(12 g.kg-1、6 g.kg-1、3 g.kg-1)。灌胃给药(除阳性药组腹腔注射外),1次/d,连续给药7 d,第7日给药后1 h;再以0.2%CCl4橄榄油溶液10 ml.kg-1剂量,腹腔注射(空白对照组除外)诱导急性肝损伤模型。造模后24 h取血处死,利用试剂盒检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)的含量;TUNUL染色法观察细胞凋亡情况;RT-PCR法检测Bcl-2及bax的基因表达;免疫组化观察肝组织bcl-2及bax的蛋白表达。结果与模型组相比,郁金各剂量组小鼠血清ALT、AST含量明显降低,TUNUL染色阳性细胞数明显减少。RT-PCR和免疫组化染色结果显示,与模型组比较郁金中、低剂量组小鼠肝组织bcl-2表达上调,bax表达下调。结论单味郁金水煎剂可抑制CCL4急性肝损伤小鼠肝细胞凋亡,其机制可能与调节肝细胞凋亡因子bcl-2/bax有关。Objective To explore effect of Curcumae on hepatocytic apoptosis in acute liver injured mice induced by CCL4.Methods The model mice of acute hepatic injury were set up by giving intraperitoneal injection of 0.2% CCl4(0.2 ml/10 mg).The serum ALT and AST were determined;Hepatocyte apoptosis and necrosis were checked with TUNE stainL.The gene expressions of Bcl2 and Bax were analyzed with RT-PCR.The protein expressions of Bcl2 and Bax were analyzed with immunohistochemistry staining.Results The serum ALT and AST in mice in model group were increased significantly compared with control group(P0.01).The serum ALT and AST in treatment group were decreased significantly compared with model group(P0.01).Fewer liver nuclei TUNEL Positive are detected in liver cells of the treatment group.Compared with model group,the gene and protein expression of bcl-2 was significantly up-regulated in liver in treatment group(P0.05),but those of bax was down-regulated(P0.05).Conclusion Curcumae could protect hepatocytes from apoptosis and necrosis in acute liver injury by ccl4 in mice,and the mechanisms in part were associated with regulations of apoptotic factors bcl-2 and bax.
