摘  要:目的 研究糖尿病小鼠学习、记忆功能及糖尿病小鼠脑海马内NT-3神经元表达的变化,并观察记忆增强肽对糖尿病小鼠学习、记忆功能及脑海马内NT-3神经元表达的影响。方法 用链脲佐菌素(streptozotocin,STZ)诱发糖尿病小鼠模型,并皮下注射记忆增强肽,4周后行水迷宫试验。水迷宫试验后对小鼠进行灌注固定,取小鼠脑海马进行冰冻切片行免疫组化染色。结果 ①记忆增强肽不影响糖尿病小鼠的血糖水平;②行为学检查(水迷宫试验)示,给予记忆增强肽的糖尿病小鼠游完全程的时间及错误反应的次数均较未给予记忆增强肽的糖尿病小鼠减少,与正常对照组相似;③神经免疫组织化学实验结果表明,记忆增强肽可阻止糖尿病小鼠海马组织内神经营养因子NT-3神经元亚群细胞表达数量的减少。结论 这些结果表明,记忆增强肽对糖尿病所致的糖尿病小鼠学习、记忆功能的减退具有改善作用,可能与海马NT-3神经元的表达正常有关。Objective To study the function of learning and memory and immunohistochemical change in NT 3 neuron expression in hippocampus of diabetic mice and to observe the protective effects of memory enhancing peptide on them.Methods Memory enhancing peptide was injected subcutaneously into streptozotocin induced diabetic mice. Four weeks later, water maze test was performed. After that, the fixative was injected intravascularly into brain and the cryostat sections were studied by immunohistochemical staining for NT 3.Results ①Memory enhancing peptide had no effcct on blood glucose of diabetic mice.②The water maze test showed that the time used to complete whole swimming journey and the number of the error behavior in diabetic mice with the injection of memory enhancing peptide were similar to the normal control group, but significantly less than those in diabetic mice without the injection.③Memory enhancing peptide increased neurotrophin 3(NT 3)expression in hippocampal tissue of diabetic mice.Conclusion Memory enhancing peptide had beneficial effects on the function of learning and memory of diabetic mice probably by means of NT 3 neuron secretion.
