摘  要:为研究LNG在大鼠原代培养肝细胞中的代谢过程,采用大鼠肝脏在体灌流技术,分离肝细胞与LNG进行原代培养,分为2.5,7.5μg/ml两组,在不同时间取0.5ml样本,用HPLC法测定LNG浓度。测定第1组K值为0.335±0.10 h^(-1),T_(1/2)为2.069±0.34h;第2组K值为0.16±0.05h^(-1),T_(1/2)为4.33±0.69h。结果显示大鼠肝细胞对LNG有一定的代谢能力,在一定的药物浓度范围内其代谢动力学符合一级动力学过程。2.069±0.34h and 4.33±0.69h-1 respectively.Conclusion; During the certain lower concerntration of LNG in cultured rat hepatocytes, LNG' smetabolism was consistent with the first order pharmakinetics.Objects; to study the metabolism of LNG in primarily cultured rathepatocytes.Methods: The hepatic perfused method in vivo was used to isolatehepatocytes, which were then cultured in RP-MI1640 medium,in whichcontained the concerntration 2.5 and 7.5 ug/ml of LNG. During thesebsequent incubation, 0.5ml sample of medium was got in various time to detect the alterations of the concerntration of LNG with HPLC method.Results; the metabolism of LNG in cultured hepatocytes was consistent with the first order pharmakinetics.The values K of both groups were 0.335 ± 0.10h-1 and 0.16 ± 0.05h-1 resperctively, and the values of Tl/2 were
