铁皮石斛多倍化诱导与鉴定研究

摘　　要：以铁皮石斛拟原球茎为材料,利用秋水仙素作为诱导剂,研究不同浓度及时间条件下,秋水仙素对铁皮石斛多倍化的诱导效果。结果表明:秋水仙素浓度为0.2%,处理时间为48h,诱导效果最佳,成活率达78%,变异率为32%;处理后的铁皮石斛在形态、气孔直径、染色体数目上都有变化,呈多倍体表型特征,其体细胞染色体数多为2n=68～76条,而对照2n=38～40条。Taking protocorm-like bodies of D.candicumas material and colchicines as mutagen,the polyploid induction of colchicines on Dendrobiumc andicumunder different concentrations of colchicines and period conditions were studied.The results showed that it resulted in the best inductive effect to use 0.2%colchicines to deal with the PLB of D.candicumof 48h.Its survival rate reached 78%and mutation rate was 32%.The plant of D.candicumtreated with colchicines was different from those of diploids in morphology,stoma diameter and chromosome numbers,showed a polyploid phenotype.Chromosome observation founded that the chromosome number of tetraploid cells was 2n=68～76,while that of diploid cells was 2n=38～40.
