保存温度与保存时间对肝脏组织中地西泮稳定性的影响

摘　　要：目的研究地西泮中毒家兔肝脏在不同温度保存时地西泮的稳定性。方法家兔以地西泮704.3 mg/kg剂量灌胃,2 h后处死,取肝脏组织,分别在室温18-24℃、冷藏4℃和冷冻-20℃的条件下保存,气相色谱/质谱联用法和气相色谱-氮磷检测器法测定不同时间点（0、1、7、14、28 d）样品中地西泮含量。结果地西泮中毒家兔肝脏在室温、冷藏和冷冻条件下保存时,肝脏中地西泮含量均呈下降趋势（P〈0.05）,3个温度组间地西泮相对含量变化差异有统计学意义（P〈0.05）。结论保存温度和时间对肝脏样品中地西泮稳定性有影响,肝脏样本在冷冻条件下保存时地西泮稳定性相对较好。Objective To study the stability of diazepam in rabbit liver stored at varying temperature conditions.Methods The rabbits were given intragastric administration of diazepam with a doze of 704.3 mg/kg and executed two hours later.The liver tissues were collected and stored at three different temperatures（room temperature,4 ℃ and-20 ℃）.The diazepam contents in rabbit liver were analyzed at selected times（0,1,7,14 and 28 d） using GC/MS and GC-NPD.Results The changes of diazepam contents in rabbit liver stored at varying temperature conditions had a tendency to decline（P〈0.05）.The significant statistical difference was shown in the change of diazepam contents in rabbit liver among different storage temperatures.Conclusion Both time and temperature have an influence on the diazepam contents in the preservative liver.The diazepam is relatively stable when liver stored at-20 ℃ condition.
