摘 要:目的探讨体外雷帕霉素(Rapa)对健康人外周血T淋巴细胞的免疫调节作用及可能机制。方法分离健康人外周血T淋巴细胞,在植物血凝素(PHA)刺激下,用不同浓度的Rapa与T淋巴细胞作用。采用MTT法检测T淋巴细胞增殖,FCM检测T淋巴细胞CD152+和CD28+的表达,RT-PCR测定IFN-γ和IL-10 mRNA的表达水平。结果 Rapa可明显抑制健康人的T淋巴细胞增殖(P<0.05),对健康人的CD3+CD152+细胞以及CD3+CD28+细胞无显著影响,Rapa可抑制健康人T淋巴细胞IFN-γ和IL-10 mRNA表达(P<0.05),并且随着Rapa浓度增加其免疫抑制作用也增强。结论 Rapa对健康人T淋巴细胞有免疫抑制作用,可能与抑制T淋巴细胞增殖和相关的细胞因子的mRNA有关。Objective To investigate the immunoregulatory effects and potential mechanism of rapamycin(Rapa) on peripheral blood T lymphocytes of healthy people in vitro. Methods Under PHA stimulating,peripheral blood T lymphocytes of normal human were treated with rapamycin. The proliferation was assessed by MTT coloremetry. FCM was used to analyze the surface markers-CD152 and CD28. The mRNA expression of IFN-γ and IL-10 were detected by real-time quantitative. Results Rapamycin cound inhibit the T lymphocytes proliferation significantly(P 0.05) ,rapamycin cound also decrease IFN-γ and IL-10 at mRNA expression(P 0.05) ,and such effect exhibited rapamycin volume dependent. There were no significant differences of CD152 expression and CD28 expression regulated by rapamycin. Conclusion Rapamycin can down-modulate function of T lymphocytes,and the potional mechanism may be related with the effect on the proliferation,and the mRNA expression of T lymphocytes differentiation-related cytokine.
